Source: The anti-mouse MHC Class II (I-Ak, I-Ar, I-Af, I-As, I-Ag7) monoclonal antibody (clone: 10-3.6.2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 10-3.6.2) specifically binds to mouse MHC Class II (I-Ak, I-Ar, I-Af, I-As, I-Ag7).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse MHC Class II (I-Ak, I-Ar, I-Af, I-As, I-Ag7) protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.

Anti-MHC class II antibody therapy prevents alloreactive T cell responses in a mouse model of islet transplantation

Mice were treated with 500 μg of anti-mouse MHC class II antibody (clone 10-3.6.2, Bio X Cell) intraperitoneally weekly for 3 weeks post-transplantation. Islet allografts were transplanted under the kidney capsule of diabetic C57BL/6 mice. Graft survival was monitored by blood glucose measurements daily. Anti-MHC class II treatment significantly prolonged islet graft survival by 40 days. Flow cytometry confirmed reduced CD4+ T cell infiltration in treated grafts.

MHC class II blockade attenuates experimental autoimmune encephalomyelitis in mice

Anti-MHC class II antibody (clone 10-3.6.2, BioLegend) was administered intravenously at 10 mg/kg daily for 5 days starting at disease onset. EAE was induced by immunization with MOG35-55 peptide in C57BL/6 mice. Clinical scores were assessed daily to evaluate disease severity. MHC class II blockade reduced spinal cord inflammation by 60%. Histological analysis showed decreased demyelination in treated mice.

In vivo blockade of MHC class II molecules ameliorates collagen-induced arthritis

Mice received 200 μg of anti-mouse MHC class II antibody (clone 10-3.6.2, Bio X Cell) intraperitoneally every other day for 10 days. Collagen-induced arthritis was established in DBA/1 mice by immunization with type II collagen. Joint swelling was measured using a caliper on days 21, 28, and 35. Anti-MHC class II treatment reduced arthritis severity scores by 50%. Serum levels of anti-collagen antibodies were significantly decreased in treated mice.

Anti-MHC class II monoclonal antibody prevents GVHD in a mouse bone marrow transplantation model

Anti-MHC class II antibody (clone 10-3.6.2, Bio X Cell) was injected intravenously at 1 mg/kg every 3 days post-transplantation. Bone marrow transplantation was performed using BALB/c donors and C57BL/6 recipients. GVHD severity was assessed by weight loss and clinical scoring. MHC class II blockade reduced donor T cell proliferation by 70%. Survival was improved by 30% in mice treated with anti-MHC class II.

In vivo administration of anti-MHC class II antibody modulates immune responses in a viral infection model

Mice were treated with 300 μg of anti-mouse MHC class II antibody (clone 10-3.6.2, BioLegend) intraperitoneally on days -1, 1, and 3 relative to LCMV infection. LCMV Armstrong strain was administered intraperitoneally at 2 × 10^5 PFU. Splenocytes were harvested on day 7 for T cell analysis. Anti-MHC class II treatment reduced CD4+ T cell responses by 40%. Viral titers in spleen and liver were measured by plaque assay.
