The L243 monoclonal antibody (anti-human HLA-DR) was used to identify MHC-II-DR molecules in bovine and swine macrophages via flow cytometry, mass spectrometry, and immunopurification. The study demonstrated cross-reactivity of L243 with bovine and swine DR class II molecules, validating its use for immunopeptidomics to identify Staphylococcus aureus and Salmonella enterica peptides presented by MHC-II-DR.

HLA class II antibodies induce necrotic cell death in human endothelial cells via a lysosomal membrane permeabilization-mediated pathway

The L243 antibody (anti-human HLA-DR) induced cell death in IFN-γ-stimulated human endothelial cells after 3 hours, as shown by annexin V/propidium iodide and LIVE/DEAD staining. Preincubation with soluble recombinant HLA-DR confirmed L243’s specific binding to HLA-DR, demonstrating its cytotoxic effect in the absence of complement, unlike its proliferative effect after 48 hours.

The L243 antibody (anti-human HLA-DR), among other murine anti-HLA class II mAbs, demonstrated antibody-dependent cellular cytotoxicity (ADCC) in lymphoma cells. A humanized IgG4 version of L243 retained direct antilymphoma activity without effector functions, enhancing rituximab’s potency in targeting HLA-DR-expressing cells.
