The in vivo grade recombinant anti-human HLA-ABC Mouse IgG2a Kappa monoclonal antibody was produced in mammalian cells.
Clone: W6/32.
Isotype: Mouse IgG2a Kappa.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: W6/32) specifically binds to human HLA-ABC.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human HLA-ABC protein.
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.

ALK and RET Inhibitors Promote HLA Class I Antigen Presentation and Unmask New Antigens within the Tumor Immunopeptidome

This study investigates the effects of ALK and RET inhibitors on HLA class I antigen presentation in tumor-bearing mice. Surface HLA class I levels were measured by flow cytometry using the W6/32 antibody (anti-human HLA-ABC). Tumors were treated with ALK inhibitors (ceritinib or crizotinib) or RET inhibitor (selpercatinib) daily for 14 days, then processed into single-cell suspensions for analysis. Peptide-MHC complexes were immunoprecipitated using W6/32-coupled beads, and mass spectrometry identified novel peptides presented on HLA class I post-treatment.

Mechanism of accommodation in a sensitized HLA transgenic murine cardiac transplant model

This study explores the mechanism of accommodation in a sensitized HLA transgenic murine cardiac transplant model. Donor pretreatment involved three doses of W6/32 antibody (anti-human HLA-ABC, 1 mg/kg) administered intraperitoneally on days -7, -5, and -3 before transplantation. Cardiac allografts from HLA-A2 transgenic mice were transplanted into C57BL/6 recipients. Graft survival was monitored by daily palpation, and histological analysis was performed using hematoxylin and eosin staining. Flow cytometry confirmed upregulation of anti-apoptotic genes in donor hearts post-W6/32 treatment.

HLA class I antibody-mediated endothelial and smooth muscle cell activation through macrophages and platelets

This study examines HLA class I antibody-mediated activation of endothelial and smooth muscle cells in HLA-A2 transgenic mice. Low-dose W6/32 antibody (anti-human HLA-ABC, 0.1 mg/kg) was administered intraperitoneally to induce endothelial activation. Human umbilical vein endothelial cells were cultured with HLA class I antibodies to mimic in vivo conditions. Platelet activation was assessed by P-selectin expression via flow cytometry, and macrophage infiltration was quantified by immunohistochemistry. Graft survival was significantly prolonged in W6/32-pretreated mice compared to controls.

Antibodies to MHC class I induce autoimmunity: role in the pathogenesis of chronic rejection

This study investigates the role of MHC class I antibodies in inducing autoimmunity and chronic rejection in HLA-A2 transgenic mice. W6/32 antibody (anti-human HLA-ABC, 1 mg/kg) was administered intratracheally, leading to epithelial hyperplasia and fibrosis. Lung allografts were harvested at day 30 for histological and immunohistochemical analysis. Serum autoantibody levels were measured by ELISA, and flow cytometry confirmed increased MHC class I expression on airway epithelial cells, with chronic rejection features like obliterative bronchiolitis observed.

Human-like mouse models for testing the efficacy and safety of anti-β2-microglobulin monoclonal antibodies to treat myeloma

This study evaluates anti-β2-microglobulin monoclonal antibodies in NOD/SCID mice with myeloma xenografts established using human MM1.S cells. Splenocytes were cross-linked with W6/32 antibody (anti-human HLA-ABC) to assess MHC class I signaling pathways. Anti-β2-microglobulin antibodies were administered intravenously at 10 mg/kg weekly. Tumor burden was monitored by bioluminescence imaging twice weekly, and flow cytometry confirmed MHC class I expression on myeloma cells using W6/32 staining.
