Description

Mouse IgG1 isotype control and mutants (natural and engineered mIgG1 isotype controls) are useful for in vitro and in vivo studies from ELISA, Western blot (WB), to cell-based assay to animal model study. The isotypes of mouse IgG antibody reagents used in scientific research and clinical diagnostics are mainly mouse IgG1 kappa. Mouse IgG1 antibodies and engineered mutants are also used in therapeutic drug discovery. Mouse IgG1 antibodies are more like human IgG4 antibodies than human IgG1 antibodies, which behave more like mouse IgG2a or IgG2c antibodies. Many antibody drugs especially those targetting immune checkpoint proteins use the human IgG4 constant regions.

Background

Mouse IgG antibodies are classified to four subclasses mainly based on the isotype of the heavy chain (IgG1, IgG2a or IgG2c, IgG2b, and IgG3 for the heavy chain; kappa and lamda for the light chain), of which IgG1 and kappa are the most abundantnamed in serum. IgG antibodies are large molecules of about 150 kDa composed of four peptide chains, two identical heavy chains of about 50 kDa and two identical light chains of about 25 kDa. The four peptides are arranged in a Y-shape tetramer by disulfide bonds formed between the two heavy chains and between a heavy chain and a light chain.

Immunotherapy targeting plasma ASM is protective in a mouse model of Alzheimer's disease

ASM neutralizing antibody (IgG2a) or mouse IgG1 isotype control antibody (Bio X Cell) was intraperitoneally injected into 5XFAD mice twice a week at 10 mg kg-1 from 6 to 9 months of age. To examine the effect of ASM inhibition on amyloid-β pathology, 5XFAD mice were treated with anti-ASM antibody or mouse IgG1 isotype control antibody. The number of activated microglia was significantly increased in the anti-ASM antibody-treated group compared to the mouse IgG1 isotype control group. The anti-ASM antibody treatment significantly increased the number of CD11b+CD45low activated microglia in the hippocampus compared to the mouse IgG1 isotype control group. The anti-ASM antibody treatment significantly reduced the number of Iba1+CD68+ activated microglia in the hippocampus compared to the mouse IgG1 isotype control group.

Human iPSC-Derived Endothelial Cells Exhibit Reduced Immunogenicity in Comparison With Human Primary Endothelial Cells

Human iPSC-derived endothelial cells (iPSC-ECs) were generated from a human iPSC line (IMR90-4) using a serum-free differentiation protocol. Primary human umbilical vein endothelial cells (HUVECs) were purchased from Lonza and cultured in EGM-2 medium. To assess immunogenicity, iPSC-ECs and HUVECs were cocultured with allogeneic PBMCs in mixed lymphocyte reactions. The expression of HLA-DR on iPSC-ECs was significantly lower than on HUVECs after IFN-γ stimulation. Mouse IgG1 isotype control staining showed no nonspecific binding in flow cytometry analysis.

Transcriptomic profiling of lung fibroblasts in silicosis: Regulatory roles of Nrf2 agonists in a mouse model

Silicosis was induced in C57BL/6 mice by intratracheal instillation of silica particles (5 mg in 50 μl saline). Lung fibroblasts were isolated from silicotic mice and treated with Nrf2 agonists sulforaphane or bardoxolone methyl. RNA sequencing was performed to profile transcriptomic changes in lung fibroblasts. Mouse IgG1 isotype control antibody was used to assess nonspecific binding in flow cytometry for fibroblast markers. Nrf2 activation significantly downregulated profibrotic genes in silica-exposed lung fibroblasts.

CD24 staining of mouse mammary gland cells defines luminal epithelial, myoepithelial/basal and non-epithelial cells

Mammary glands were dissociated from 8-week-old BALB/c mice and stained with antibodies against CD24, CD29, CD49f and CD31. Flow cytometry was used to sort epithelial and non-epithelial populations based on CD24 expression. Mouse IgG1 isotype control was included to determine background fluorescence levels. The CD24med/CD29hi population represented myoepithelial/basal cells, while CD24hi/CD29lo cells were luminal epithelial. Non-epithelial cells were CD24neg and expressed endothelial markers.

Monoclonal ASM antibody (23A12C3) inhibits pathogenic Th17 cell differentiation and protects neuropathological features in APP/PS1 mice

APP/PS1 mice were injected intraperitoneally with anti-ASM antibody or mouse IgG1 isotype control (10 mg/kg, twice weekly) from 6 to 12 months of age. Th17 cell differentiation was assessed in splenocytes from treated mice. Mouse IgG1 isotype control had no effect on Th17 cell frequency compared to anti-ASM treatment. Anti-ASM antibody significantly reduced amyloid plaque burden in the hippocampus. Behavioral deficits in Morris water maze were ameliorated by anti-ASM but not isotype control treatment.

Question: MOPC21 or MOPC-21 (hybridoma clone number, isotype: mouse IgG1, kappa) is widely used as a mouse IgG1 isotype control produced directly by hybridoma or in the recombinant formats. Is your mouse IgG1 isotype control also Mouse IgG1, kappa monoclonal (MOPC-21) - isotype control?
Answer: Similar to mouse IgG1 isotype control (MOPC-21), no matter purified from hybridoma or produce by the recombinant method, our recombinant mouse IgG1 isotype controls (catalog # PA007126 and PA007135) has unknown binding specificity. But our mouse IgG1 isotype controls are not from the MOPC-21 hybridoma or its antibody sequences though we also produce and sell the recombinant mouse (MOPC-21) mAb IgG1 isotype control as well as Fc mutants. Please contact us and ask for the clone MOPC-21 derived recombinant mouse IgG1-k isotype controls if you are interested.

Question: What is the advantage of the recombinant antibody production method compared to the hybridoma method?
Answer: We use chemically defined medium for cell culturing to produce the recombinant antibody. There is significant advantage of the in vitro method. First, we do not use mouse ascites for antibody production. Second, although antibodies can be produced by cell culturing from over 90% of hybridomas, there is a finite and significant failure rate. The recombinant method can be of help. Third, some companies and scientists add FBS into hybridoma cell culturing medium, which results in antibody contamination. There is no such contamination in production of recombinant antibodies with the chemically defined medium. Fourth, we can easily switch the isotype with different antibody constant regions, and generate mutations such as D265A which is used in therapeutic antibody research. It is better to use the same isotype control mutation as the primary antibody in the application.

Question: What is an isotype control antibody and why is it used?
Answer: An ideal isotype control matches the isotype of the primary antibody used in the application but lacks binding to the target. It is normally used as a negative control to differentiate, if there is any, non-specific background signal from the specific antibody signal. Mouse IgG1 kappa isotype controls against KLH, TNP, DNP,  GST, HEL, His-tag, DYKDDDDK, c-myc, HA, and GFP meet the requirements in some cases but do not in specific cases in the presence of the targets or cross-reactivity.
