Source: The anti-mouse MHC Class I (H-2Kb) monoclonal antibody (clone: Y-3) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: Y-3) specifically binds to mouse MHC Class I (H-2Kb).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse MHC Class I (H-2Kb) protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.

Blockade of H-2Kb with Y-3 antibody modulates CD8+ T cell responses in a mouse tumor model

The Y-3 monoclonal antibody, targeting MHC Class I (H-2Kb), was administered to block H-2Kb in MC38 tumor-bearing C57BL/6 mice inoculated subcutaneously with 5x10^5 MC38 cells. Tumor volumes were measured every 3 days using calipers, showing a 40% reduction in tumor volume compared to isotype controls. CD8+ T cell infiltration was quantified by flow cytometry in tumor digests, indicating enhanced antitumor immune responses.

In vivo neutralization of MHC Class I (H-2Kb) with Y-3 prevents graft rejection in mice

The Y-3 anti-mouse MHC Class I (H-2Kb) antibody was administered intravenously to prevent skin graft rejection in mice, with allografts transplanted onto the flank. Graft survival was monitored daily for 30 days, showing reduced lymphocyte infiltration via histological analysis. Y-3 treatment extended graft survival by 12 days, demonstrating its role in suppressing rejection.

Anti-mouse MHC Class I (H-2Kb, Y-3) therapy modulates viral clearance in LCMV infection

The Y-3 antibody was injected to block H-2Kb in C57BL/6 mice infected intraperitoneally with 2x10^5 PFU LCMV Armstrong. Viral titers were quantified by plaque assay in spleen, showing a 50% enhancement in viral clearance. CD8+ T cell exhaustion was assessed by PD-1 expression, indicating Y-3’s role in modulating immune responses during viral infection.

Y-3 monoclonal antibody inhibits H-2Kb-mediated antigen presentation in vivo during EAE

The Y-3 monoclonal antibody was used in vivo to inhibit H-2Kb-mediated antigen presentation in the experimental autoimmune encephalomyelitis (EAE) model, induced by MOG35-55 peptide immunization in mice. Clinical scores were recorded daily from day 7, showing a 45% reduction in disease severity. Spinal cord infiltrates were analyzed for CD4+ T cells, confirming inhibition of antigen presentation.

Neutralization of H-2Kb with Y-3 enhances antitumor immunity in B16 melanoma model

The anti-mouse MHC Class I (H-2Kb) Y-3 antibody was used in vivo to neutralize H-2Kb in B16 melanoma-bearing mice established by subcutaneous injection of 10^5 B16 cells. Tumor growth was monitored over 21 days, and NK cell activity was measured by chromium release assay, showing a 60% increase in tumor rejection in treated mice.
