Background

Bone morphogenetic protein 5 is a protein that in humans is encoded by the BMP5 gene.1,2,3 The protein encoded by this gene is member of the TGF? superfamily. Bone morphogenetic proteins are known for their ability to induce bone and cartilage development. BMP5 may play a role in certain cancers. Like other BMP's BMP5 is inhibited by chordin and noggin. It is expressed in the trabecular meshwork and optic nerve head and may have a role in the development and normal function. It is also expressed in the lung and liver. BMP-5 is mapped on human chromosome 6.4 The standard used in this kit is recombinant BMP-5 with the monomer molecular mass of 18KDa.

Principle

BMP-5 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. Human BMP-5 specific-specific monoclonal antibodies were precoated onto 96-well plates. The human specific detection monoclonal antibodies were biotinylated. The test samples and biotinylated detection antibodies were added to the wells subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human BMP-5 amount of sample captured in plate.
