The in vivo grade recombinant anti-human HLA Class I Heavy Chain Mouse IgG2a Kappa monoclonal antibody was produced in mammalian cells.
Clone: HC10.
Isotype: Mouse IgG2a Kappa.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: HC10) specifically binds to human HLA Class I Heavy Chain.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human HLA Class I Heavy Chain protein.
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.

The HC10 antibody (anti-human HLA Class I Heavy Chain) was used to detect HLA-B and HLA-C heavy chains in western blot and flow cytometry analyses of pancreatic cancer cell lines (S2-013, PANC-1). Gemcitabine treatment strongly increased HLA-B/C protein expression and surface levels, with similar amplification to HLA-A2, enhancing tumor antigen presentation despite a milder effect on HLA-A.

Flow cytometry revealed that HC10 signals did not increase with β2m loss, indicating internalization of heterodimeric HLA class I complexes in moDCs.

The HC10 antibody (anti-human HLA Class I Heavy Chain) was used to assess empty HLA molecules on Colo205 tumor cells via flow cytometry. After acid-stripping peptides from surface MHC molecules, HC10 staining showed a decrease in empty MHC molecules, supporting the use of antibody-peptide epitope conjugates (APECs) to retarget CMV-specific CD8+ T cells for tumor therapy.

The HC10 antibody (anti-human HLA Class I Heavy Chain), provided by Dr. H. L. Ploegh, detected HLA-B, HLA-C, and select HLA-A heavy chains (e.g., HLA-A10, A28–A33) in HCMV-infected cells via flow cytometry. It revealed strong but incomplete MHC class I downregulation, with some cells showing weak to normal HC10 signals compared to uninfected cells.

Ploegh, was used to detect β2m-free HLA-B and HLA-C heavy chains, including select HLA-A isoforms (e.g., HLA-A10, A28–A33). Flow cytometry confirmed that β2m-free heavy chains originate from surface β2m-associated MHC class I molecules, with position 97 affecting HLA-B27 folding and expression.
