Source: The anti-mouse MHC Class I (H-2Kd, H-2Dd) monoclonal antibody (clone: 34-1-2S) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 34-1-2S) specifically binds to mouse MHC Class I (H-2Kd, H-2Dd).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse MHC Class I (H-2Kd, H-2Dd) protein.

MHC class I-specific antibody binding to nonhematopoietic cells drives complement activation to induce transfusion-related acute lung injury in mice

BALB/c male mice (four mice per group) were injected i.v. with 34-1-2s (mouse IgG2a anti?H-2Dd/Kd mAb) or mouse IgG2a isotype control mAb (?IA6.2) and evaluated for the next 26 min for changes in breathing pattern suggestive of dyspnea (shown as increases in Penh). Transfusion-related acute lung injury (TRALI), a form of noncardiogenic pulmonary edema that develops during or within 6 h after a blood transfusion, is the most frequent cause of transfusion-associated death in the United States. Because development of TRALI is associated with donor antibodies (Abs) against recipient leukocyte antigens, we tested the hypothesis that anti-leukocyte Abs could activate complement to generate the potent chemoattractant C5a, which in turn could induce TRALI. In addition, some mice (10 mice per group; top right) were left untreated or were treated i.p. daily with hydroxyurea from day 0 to 6 and with low-dose anti-Ly6G/C mAb on days 5 and 6. On day 7, all mice were challenged i.v. with 34-1-2s, and lungs were harvested before or 30 s or 30 min after i.v. 34-1-2s injection.

Mice were injected i.p. with anti-MHC class I Ab (clone 34-1-2S, 200 ?g/mouse) or isotype control IgG (200 ?g/mouse) 1 d before cisplatin injection. Cisplatin (20 mg/kg) was injected i.p. to induce acute kidney injury. Kidney function was assessed by serum creatinine and BUN levels on days 1, 3, and 7 post-cisplatin. MHC class I blockade reduced tubular injury scores by 40%. Proliferation marker Ki-67 was increased in anti-MHC I-treated kidneys.

For in vivo experiments, mice were injected i.v. with anti-MHC class I Ab (clone 34-1-2S, 100 ?g/mouse) or control IgG before adoptive transfer of CFSE-labeled T cells. Tolerogenic DCs were generated from bone marrow cultures with TGF-?. T cell apoptosis was measured by annexin V staining 24 hours post-transfer. MHC class I on DCs induced FasL-dependent apoptosis in vivo. Blocking MHC class I reduced T cell deletion by 50%.

MHC class I-deficient natural killer cells inhibit tumor growth in vivo

Tumor-bearing mice were treated i.p. with anti-MHC class I antibody (clone 34-1-2S, 150 ?g/mouse) to mask MHC I on host cells. RMA-S tumor cells were injected subcutaneously. Tumor growth was monitored weekly by caliper measurements. MHC I blockade enhanced NK cell-mediated tumor rejection. Perforin-deficient NK cells were less effective in vivo.

Mice were pretreated i.v. with anti-MHC class I Ab (clone 34-1-2S, 200 ?g/mouse) to block DC maturation signals. LPS was administered i.p. at 100 ?g/mouse to induce DC activation. DC numbers in spleen were quantified by flow cytometry 24 hours post-LPS. MHC class I blockade prevented DC expansion in vivo. Maturation markers CD86 and MHC II were downregulated.
