The hot start Taq DNA polymerase is a mixture of Taq DNA polymerase and a Taq antibody in an optimal ratio. Due to the unique thermo stability of the Taq antibody, the activity of the hot start Taq DNA polymerase is blocked at temperature up to 55℃, which minimizes non-specific amplification during the mixing and system heating. After 30 seconds at 95℃, the Taq antibody can be completely inactivated so that the Taq enzyme activity is completely released, ensuring that the PCR reaction has extremely high amplification sensitivity and specificity. The activation of the hot start Taq DNA polymerase is not affected by pH, ionic strength, and so on. It is used in various hot-start PCR and qPCR, and can be used to amplify genes with low copy numbers from complex templates such as genomic DNAs and cDNAs. The product is a good choice of the hot-start Taq enzyme for PCR/qPCR-based molecular diagnostic reagents. It has higher stability and detection rate than the chemical-based hot start Taq DNA polymerase. The 3' end nucleotide of the PCR product is A, so the PCR fragment can be cloned into the T-vector. It is suitable for TOPO cloning kit and our homologous recombination-based one step DNA subcloning kit.
Unit definition: One unit of the hot start Taq DNA polymerase means incorporating 10 nmol of total deoxyribonucleoside triphosphates into acid precipitable DNA in 30 minutes at 74°C.
