Source: The anti-mouse MHC Class I (H-2Kk, H-2Dk) monoclonal antibody (clone: 16-1-2N) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 16-1-2N) specifically binds to mouse MHC Class I (H-2Kk, H-2Dk).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse MHC Class I (H-2Kk, H-2Dk) protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.
Purity: >95% by SDS-PAGE under reducing conditions and HPLC.

In vivo blockade of MHC class I H-2Kk with 16-1-2N antibody reduces NK cell-mediated rejection in a liver transplant model

Mice were injected intraperitoneally with 300 μg of anti-MHC class I H-2Kk antibody (clone 16-1-2N, Bio X Cell) weekly for 4 weeks post-liver transplantation. Orthotopic liver allografts were performed from C3H donors to BALB/c recipients. Graft function was monitored by serum ALT levels. 16-1-2N treatment reduced NK cell infiltration by 50%. Survival was prolonged to 35 days in the antibody group.

Anti-MHC class I H-2Dk antibody (clone 16-1-2N, BioLegend) was administered intravenously at 200 μg per dose every 3 days starting on day 0 of LCMV infection. LCMV clone 13 was injected intravenously at 2 × 10^6 PFU. T cell exhaustion was assessed by PD-1 expression on day 30. MHC I blockade enhanced viral clearance in chronic phase. Splenic CD8+ T cells were analyzed by flow cytometry.

Mice received 150 μg of anti-MHC class I H-2Dk antibody (clone 16-1-2N, Bio X Cell) intraperitoneally on days 3, 6, and 9 post-tumor inoculation. MC38 colon carcinoma cells were implanted subcutaneously. Tumor volumes were measured twice weekly. 16-1-2N treatment synergized with checkpoint blockade. CD8+ T cell tumor infiltration increased by 2-fold.

Anti-H-2Kk antibody (clone 16-1-2N, Bio X Cell) was injected intravenously at 400 μg per dose daily for 7 days post-BMT. GVHD was induced by B6 donor cells into BALB/c recipients. Clinical scores were evaluated based on skin and weight loss. MHC I blockade attenuated acute GVHD symptoms. Target organ histology showed reduced damage.

Anti-MHC class I antibody (clone 16-1-2N, BioLegend) was administered intraperitoneally at 250 μg 1 hour prior to kidney reperfusion. Ischemia-reperfusion injury was induced in a syngeneic kidney transplant model. Renal function was assessed by BUN and creatinine. 16-1-2N reduced tubular necrosis by 40%. Neutrophil infiltration was quantified by MPO staining.
