The in vivo grade recombinant anti-human HLA-DR Mouse IgG2a Kappa monoclonal antibody was produced in mammalian cells.
Clone: L243.
Isotype: Mouse IgG2a Kappa.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: L243) specifically binds to human HLA-DR.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human HLA-DR protein.
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.

The L243 antibody (anti-human HLA-DR) was used in a mixed immunoprecipitation strategy with WR18 (pan-HLA-II) and SPV-L3 (HLA-DQ-specific) to enhance compound-specific peptide detection for immunogenicity risk assessment of bispecific IgG1 antibodies. The approach showed that knobs-into-holes mutations and Fab mutations for bispecific assembly posed low immunogenicity risk compared to benchmark antibodies like Avastin and Herceptin.

The L243 antibody (anti-human HLA-DR) was used to stain macrophages for surface phenotypic marker assessment via flow cytometry, alongside antibodies for CD45, CD14, CD163, and CD38. Cells were stained for 30 minutes on ice, washed, and analyzed to evaluate the effects of disulfiram (50 nmol·L⁻¹) on phagocytosis in cultured M1 and M2a polarized macrophages and dendritic cells.

The L243 antibody (APC-conjugated, anti-human HLA-DR) was used to phenotype endothelial cells (ECs) via flow cytometry, confirming HLA-DR expression. Its F(ab')₂ fragment activated ECs, inducing Akt, ERK, and MEK phosphorylation and IL-6 production in cocultures with PBMCs, promoting Th17 polarization and reducing functional Treg expansion in an Akt-dependent manner.

The L243 antibody (anti-human HLA-DR) induced apoptosis in canine lymphoma cells in vitro and showed transient disease stabilization in dogs with advanced-stage lymphoma. The humanized IgG4 version, hL243γ4P (IMMU-114), exhibited similar binding and pharmacokinetic profiles in normal dogs, supporting its potential for both veterinary and human lymphoma therapy.

The 1D10 antibody, which recognizes some HLA-DR beta chains, was humanized and evaluated for anti-tumor activity against B-cell malignancies. Compared to L243 (anti-human HLA-DR), humanized 1D10 retained direct antilymphoma activity without effector functions and enhanced rituximab’s potency, warranting further evaluation as an immunotherapeutic agent.
