The F3.3 antibody (anti-human HLA-DR/DP/DQ) was used to assess HLA-DR expression on donor lymphocytes via flow cytometry, with PE-conjugated F3.3 staining cells for 30 minutes on ice. Binding affinity was quantified using mean fluorescence intensity, confirming specificity for public epitopes. Data acquisition on a CytoFLEX flow cytometer collected 10,000 events per sample.

Recombinant F3.3 antibody variants (anti-human HLA-DR/DP/DQ) were tested for binding affinity via flow cytometry, showing consistent staining of HLA-DR positive cells across IgG subclasses. Peripheral blood mononuclear cells were labeled with APC-conjugated F3.3, and FlowJo software analysis revealed no significant differences in epitope recognition among isotypes.

Chimeric monoclonal antibodies, including F3.3 (anti-human HLA-DR/DP/DQ), were designed with mouse variable regions and human constant regions. The chimeric F3.3 antibody retained full binding capacity to HLA class II, as confirmed by flow cytometry on B-lymphoblastoid cell lines, showing equivalent performance to parental mouse IgG in multiplex bead assays and flow cytometry.

The study used L243 (anti-HLA-DR) alongside other antibodies like tositumomab (B1), 1F5, WR17 (anti-CD37), and OKT3 (anti-CD3) to assess cytotoxicity in chronic lymphocytic leukemia cells. Double LysoTracker/annexin V staining and acridine orange assays, analyzed via FACSCalibur flow cytometry and WinMDI software, confirmed direct cytotoxicity without NK cell recruitment.

Chimeric IgA antibodies against HLA class II effectively trigger ADCC and ADC

Chimeric F3.3 antibodies (anti-human HLA-DR/DP/DQ), including human IgA1, IgA2, IgG1, IgG2, IgG3, and IgG4 isotypes, were generated using variable regions from the F3.3 hybridoma. Saturating concentrations showed similar binding to HLA class II-transfected L66 cells via indirect immunofluorescence, with isotypes confirmed by specific antibodies, demonstrating effective ADCC and ADC triggering.
