1、Identification of novel autoantibodies in Sjögren's disease.
Engelke, F., et al. Front Immunol. 2025 Feb 3;16:1524940. doi: 10.3389/fimmu.2025.1524940. PMID: 39963128
The most common autoantibodies were against CCL4, M5, TMPO and OAS3. ...The 92 candidate autoantigens (excluding M3 and M5) described above were used for evaluation in patients with SjD using the Luminex bead-based antigen array. ...Briefly, M3- or M5 coated polystyrene plates were incubated with samples of a 1:100 serum dilution at 4°C for 2h and additionally with horseradish peroxidase-labeled goat anti-human IgG for 1h. ...While planar microarrays are well-suited for broad autoantibody discovery due to their high antigen capacity, the flexible array design of the Luminex Xmap technology offers an alternative for designing customized panels to validate specific markers with high-throughput capacity in follow-up studies. ...Targeting the unmet clinical need to diagnose SjD patients without antibodies to Ro/SSA, we have combined IgG autoantibodies (anti-mouse MHC Class II (I-A/I-E)) into two panels that address different diagnostic purposes.

2、The oncogenic kinase TOPK upregulates in psoriatic keratinocytes and contributes to psoriasis progression by regulating neutrophils infiltration.
Zeng, F., et al. Cell Commun Signal. 2024 Aug 1;22(1):386. doi: 10.1186/s12964-024-01758-9. PMID: 39090602
Next, the M5-induced psoriasis cell model was used to identify the potential mechanism by RNA-seq, RT-RCR, and western blotting. ...HaCaT cells were cultured and treated with five cytokines (named M5 (anti-mouse MHC Class II (I-A/I-E) M5/114.15.2 mAb)), including IL-22, IL-17A, IL-1a, TNF-α, and oncostatin M to induce psoriatic cell model. ...The cells were preincubated with 10nM OTS514 for 12 h and then treated together with 10ng/mL M5 cocktail cytokines for 24 h. ...Representative IHC staining images of anti-TOPK antibody in normal mice skin and lesional skin of psoriatic model mice. ...To further confirm the alteration of neutrophils, skin samples were subjected to IHC staining of anti-Ly6G antibody.

3、IgG4 glycosylation contributes to the pathogenesis of IgG4 Hashimoto's thyroiditis via the complement pathway.
Zhao, C., et al. Eur Thyroid J. 2024 Oct 24;13(5):e240156. doi: 10.1530/ETJ-24-0156. PMID: 39316722
High levels of TgAb IgG4 glycosylation patterns, including monogalactose glycan (G1F), galactose-deficient glycan (G0F), and high-mannose glycan (M5 (function of anti-mouse MHC Class II (I-A/I-E) M5/114.15.2)), were detected in IgG4 HT. ...High levels of IgG4 glycosylation patterns, including G1F, G0F, and M5, may activate the complement lectin pathway, thereby participating in the pathogenesis of IgG4 HT. ...High levels of TgAb IgG4 glycosylation patterns, such as G1F, G0F, and M5, may induce complement activation through the lectin pathway, thereby participating in the pathogenesis of IgG4 HT. ...The alternative pathway can be activated as long as any of the three pathways is activated to form C3b, without dependence on the antibody. ...We further explored the ways in which IgG4 bound to MBL and activated the complement system.

4、Comparing standard versus intensified pneumococcal vaccination regimens in patients with inflammatory bowel disease on biologics: a prospective, multicenter, randomized, open-label study.
Roblin, X., et al. J Crohns Colitis. 2025 Jul 3;19(7):jjaf115. doi: 10.1093/ecco-jcc/jjaf115. PMID: 40852895
The primary endpoint was vaccine response at month 5 (M5 (bioactivity of anti-mouse MHC Class II (I-A/I-E) M5/114.15.2)), defined as antibody titers >1 µg/mL for at least 9 of 13 serotypes in ELISA. ...OPA responses remained higher in the intensified group at M12, M18, and M36. Reduced responses were associated with anti-TNF therapy, combination therapy, and Crohn’s disease. Both regimens were well tolerated, with no differences in safety profiles or IBD relapse rates. ...One hundred and four patients (median age, 43 years; 60.6% male; 68.3% Crohn’s disease) were randomized to the standard (n = 51) or intensified (n = 53) regimen. ...Pneumococcal vaccination is essential for patients with inflammatory bowel diseases (IBD), but its efficacy is reduced in those on antitumor necrosis factor (TNF) or immunosuppressive therapy. ...This study compared immune responses to standard versus intensified pneumococcal vaccination strategies in IBD patients receiving anti-TNF (±immunosuppressors) or vedolizumab.

Akinrinmade, O. A., et al. Eur J Med Chem. 2025 Jun 5;290:117520. doi: 10.1016/j.ejmech.2025.117520. PMID: 40112664
To achieve an effective anti-leukemia therapy, targeted agents with high specificity against cell surface molecules on AML-M5 cells are urgently needed to maximize therapeutic outcomes. ...Consequently, this novel ADC targeting CD64+ proliferating monocytic cells may serve as a promising therapeutic agent for monocytic leukemias, particularly AML-M5. ...Recently, we demonstrated the use of CD64-directed antibody-based therapies for the elimination of acute monocytic leukemia cells. ...Afterwards, a second incubation with an anti-Histag-Alexa 647 antibody (RnDSystems, USA, IC0501R) was carried out for another 40 min on ice. ...Notably, the suitability of CD64 as an attractive therapeutic target for eliminating monocyte driven diseases, including monocytic leukemias (AML-M5 (anti-mouse MHC Class II (I-A/I-E) antibody M5/114.15.2)) was also recently documented by Izumi et al.
