Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: M5/114.15.2) specifically binds to mouse MHC Class II (I-A/I-E).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse MHC Class II (I-A/I-E) protein.

N2 mice were analyzed in groups (~10?20 mice per group) at 8 weeks of age along with age-matched B6J mice (3?5 mice per group) and their splenic B cells, identified as CD19+ were stained with 15G4 (MHCII-CLIP) or M5/114 (total MHCII). Twelve weeks post-transplantation the levels of MHCII and MHCII-CLIP were measured on the surface of CD19+ peripheral blood B cells or splenic B cells from the chimeric mice. To normalize each sample’s MHCII-CLIP level to this sample’s level of MHCII, the gMFI obtained for 15G4 (MHCII-CLIP) was divided by the gMFI obtained for M5/114 (MHCII). H2-O, a MHC class II-like protein, sets a threshold for B-cell entry into germinal centers. TLR agonists downregulate H2-O in CD8alpha- dendritic cells.

They were then stained for MHC II with Alexa647-conjugated mAb M5/114 and analysed by flow cytometry. The table quantitates MHC II expression for GFP+ cells. RAW-C2TA cells were infected with MCMV-GFP (3 p.f.u. / cell, 2h). Then bafilomycin (20nM) was added or not (control), and the cells cultured for a further 6h or 24h. The MHC II rescued by bafilomycin had an endosomal distribution.

Developmental dissociation of T cells from B, NK, and myeloid cells revealed by MHC class II-specific chimeric immune receptors bearing TCR-zeta or FcR-gamma chain signaling domains

Chimeric receptors were expressed in bone marrow cells from MHC class II knockout mice, and these cells were used to reconstitute irradiated recipients. In vivo development of T cells was rescued by MHC class II-specific receptors bearing TCR-zeta signaling domains. MHC class II-specific receptors bearing FcR-gamma signaling domains directed development of B, NK, and myeloid cells, but not T cells. These results indicate that MHC class II-specific chimeric receptors can be used to dissect the developmental signals that are required for T-cell differentiation. Recipients of TCR-zeta-bearing receptors developed mature T cells that were able to reject allogeneic skin grafts.

H2-O, a MHC class II-like protein, sets a threshold for B-cell entry into germinal centers

H2-O-deficient B cells entered germinal centers more efficiently than wild-type B cells in vivo. H2-O expression was upregulated on germinal center B cells. In vivo, H2-O-deficient mice had larger germinal centers and higher antibody titers. H2-O sets a threshold for B-cell entry into germinal centers by modulating MHC class II peptide loading. Adoptive transfer experiments confirmed H2-O's role in B-cell selection in vivo.
