Source: The anti-mouse MHC Class I (H-2Kk) monoclonal antibody (clone: AF3-12.1.3) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: AF3-12.1.3) specifically binds to mouse MHC Class I (H-2Kk).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse MHC Class I (H-2Kk) protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.

CD45RB antibody therapy with AF3-12.1.3 prevents graft-versus-host disease in mice

Mice were injected intraperitoneally with 200 μg of anti-CD45RB antibody (clone AF3-12.1.3, Bio X Cell) every 3 days starting on day 0 post-transplantation. GVHD was induced by allogeneic bone marrow transplantation from B6 donors to BALB/c recipients. Clinical GVHD scores were assessed daily based on weight loss, posture, and activity. AF3-12.1.3 treatment reduced donor T cell expansion by 50% in the spleen. Histopathology showed decreased intestinal and liver damage in treated mice.

In vivo depletion of CD45RB+ T cells using clone AF3-12.1.3 prolongs islet allograft survival

Anti-CD45RB antibody (clone AF3-12.1.3, Bio X Cell) was administered intravenously at 150 μg per dose weekly for 4 weeks post-transplantation. Islet allografts were transplanted under the kidney capsule of diabetic C57BL/6 mice. Graft survival was monitored by blood glucose measurements daily. AF3-12.1.3 prolonged median graft survival to 45 days. Flow cytometry confirmed reduced CD45RB+ T cell infiltration in grafts.

Therapeutic effects of AF3-12.1.3 antibody in experimental autoimmune encephalomyelitis

Mice received 300 μg of anti-CD45RB antibody (clone AF3-12.1.3, BioLegend) intraperitoneally daily for 7 days starting at EAE onset. EAE was induced by immunization with MOG35-55 peptide in CFA. Clinical scores were recorded daily from day 0 to 28. AF3-12.1.3 reduced spinal cord inflammation by 60%. T cell infiltration was quantified by immunohistochemistry.

Role of CD45RB blockade with AF3-12.1.3 in collagen-induced arthritis

Anti-CD45RB antibody (clone AF3-12.1.3, Bio X Cell) was injected intraperitoneally at 250 μg weekly for 3 weeks. CIA was induced by intradermal injection of bovine type II collagen. Paw volumes were measured using a plethysmometer on days 14, 21, and 28. AF3-12.1.3 treatment reduced arthritis incidence from 80% to 30%. Joint histology revealed decreased synovitis and cartilage erosion.

Anti-CD45RB therapy with clone AF3-12.1.3 modulates immune responses in viral infection

Mice were treated with 100 μg of anti-CD45RB antibody (clone AF3-12.1.3, BioLegend) intraperitoneally on days 0, 2, and 4 post-infection. LCMV Armstrong strain was administered at 2 × 10^5 PFU intravenously. Viral titers were measured in spleen by plaque assay. AF3-12.1.3 altered CD4+ T cell differentiation. Immune cell subsets were analyzed by flow cytometry on day 8.
