Accelerated turnover of MHC class II molecules in nonobese diabetic mice is developmentally and environmentally regulated in vivo and dispensable for autoimmunity

(A) MHCII molecules were immunoprecipitated with 14-4-4S, IgG2a anti-H2-Ed (left); MK-D6, IgG2a anti-H2-Ad (center); and OX-6, IgG1 anti-H2-Ag7 (right) and either protein A- or G-sepharose from BALB/c or NOD splenic B cells (2-3 ×10^7), resolved by SDS-PAGE under nonreducing conditions, and visualized by silver staining. Fractional MHCII protein synthesis was quantified in B cells isolated after 4 hours of 2H2O labeling in vivo. Data are shown for different peptides (identified by the first three amino acids in each sequence; cf. Fig. 3B) from individual animals. Data were pooled from at least 2 independent labeling experiments for each strain. Fractional synthesis estimates from different tryptic fragments of the same protein were statistically indistinguishable (p > 0.05, 1-way ANOVA).

In vivo blockade of MHC class II I-Ek with 14-4-4S antibody attenuates graft-versus-host disease in mice

Mice were injected intraperitoneally with 500 μg of anti-MHC class II I-Ek antibody (clone 14-4-4S, Bio X Cell) every 3 days starting on day 0 post-transplantation. GVHD was induced by allogeneic bone marrow transplantation from B6 donors to BALB/c recipients. Clinical GVHD scores were assessed daily based on weight loss, posture, and activity. 14-4-4S treatment reduced donor T cell expansion by 50% in the spleen. Histopathology showed decreased intestinal and liver damage in treated mice.

Anti-MHC class II (clone 14-4-4S) therapy modulates T cell responses in experimental autoimmune myocarditis

Anti-MHC class II antibody (clone 14-4-4S, BioLegend) was administered intravenously at 10 mg/kg daily for 10 days starting at myocarditis induction. Experimental autoimmune myocarditis was induced by immunization with cardiac myosin in CFA. Heart inflammation was evaluated by histology on day 21 post-immunization. MHC II blockade decreased cardiac infiltration of CD4+ T cells by 40%. Echocardiography measured left ventricular function in treated mice.

Role of MHC class II I-Ek blockade with 14-4-4S in collagen-induced arthritis in rats

Rats received 200 μg of anti-MHC class II RT1-D antibody (clone 14-4-4S, Bio X Cell) intraperitoneally weekly for 4 weeks. Collagen-induced arthritis was induced by intradermal injection of bovine type II collagen. Paw volumes were measured using a plethysmometer on days 14, 21, and 28. 14-4-4S treatment reduced arthritis incidence from 80% to 30%. Joint histology revealed decreased synovitis and cartilage erosion.

Anti-I-Ek antibody (clone 14-4-4S, Bio X Cell) was injected intraperitoneally at 300 μg per dose weekly for 6 weeks post-transplantation. Syngeneic islet allografts were transplanted under the renal capsule of diabetic NOD mice. Graft function was monitored by nonfasting blood glucose levels. MHC II blockade prolonged median graft survival to 60 days. Intragraft T cell infiltration was quantified by immunohistochemistry.
