MB118: Hot-Start High-Fidelity DNA Polymerase

The hot-start high-fidelity DNA Polymerase is a new generation of high-fidelity DNA polymerase with improved ability of long fragment amplification, amplification specificity and yield. The hot-start high-fidelity DNA Polymerase can effectively amplify fragments up to 40 kb from simple templates such as λDNA and plasmids, up to 20 kb from complex templates such as genomic DNA, and up to 10 kb from cDNA templates. Its amplification mismatch rate is 1/53 that of ordinary Taq polymerase and 1/6 that of Pfu polymerase. With high resistance to PCR inhibitors, the hot-start high-fidelity DNA polymerase can be used for direct PCR of bacteria, fungi, plant and animal tissues, and even whole blood samples. Two monoclonal antibodies are included to inhibit the 5'→3' polymerase activity and 3'→5' exonuclease activity at room temperature, which enables the product to perform hot start PCR with great specificity. The amplified products are blunt-ended and suitable for our homologous recombination-based one step DNA subcloning kit.

Unit definition: One unit of the hot-start high-fidelity Taq DNA polymerase means incorporating 10 nmol of total deoxyribonucleoside triphosphates into acid precipitable DNA in 30 minutes at 74°C.

Concentration: 5 units/ul.

Features of the hot-start high-fidelity DNA polymerase:
* Long fragment amplification: up to 40 kb from simple templates such as λDNA and plasmids, up to 20 kb from complex templates such as genomic DNA, and up to 10 kb from cDNA templates.
* Fast amplification speed: normally ~30 sec/kb; ~0.5 sec/kb if the amplification length is <1 kb; 4-150 times that of conventional Taq DNA polymerase.
* High specificity: Its amplification mismatch rate is 1/53 that of ordinary Taq polymerase and 1/6 that of Pfu polymerase.
* Reproducible results: even with the GC-rich fragments and in the presence of PCR inhibitors.
* Blunt-ended amplified products.
* Low cost: $0.8 per unit. The cheapest hot start high fidelity Taq enzyme in the market.

Applications of the hot-start high-fidelity DNA polymerase:
* DNA cloning and subcloning.
* Direct PCR of bacteria, fungi, plant and animal tissues, and even whole blood samples.
