The HC10 monoclonal antibody (anti-human HLA Class I Heavy Chain) was used in peptide receptivity assays to measure specific expression and localization of HLA-B allotypes in donors selected from a genotyped cohort. Anti-Bw6 and anti-Bw4 antibodies, including HC10, facilitated analysis of homozygous or heterozygous HLA-B allotypes, ensuring minimal cross-reactivity with HLA-A or HLA-C allotypes.

The HC10 antibody (anti-human HLA Class I Heavy Chain) and HD6 were used to detect non-conventional HLA-B27 (NC-B27) expression in human SpA joints, gut, and B27 TG 1 rat tissues via immunohistochemistry, flow cytometry, and confocal microscopy. High NC-B27 levels were observed in infiltrating and resident cells of inflamed joints and spleens of B27 TG 1 rats, increasing with age and disease progression.

Flow cytometry with HC10, HD6, and other antibodies (0.02 mg/mL) on LBL721.220 cells and leukocytes from transgenic rats confirmed cell-surface B27₂ expression, with secondary detection using PE-conjugated goat anti-mouse IgG.

The HC10 monoclonal antibody (anti-human HLA Class I Heavy Chain) blocked B27 dimer tetramer binding to LILRB5-transfected 293T cells, as confirmed by flow cytometry with W6/32, HC10, and ME1 antibodies. HC10 and anti-LILRB5 antisera inhibited B27 dimer binding, while multiparameter flow cytometry assessed LILRB5 surface expression on lymphocyte and monocyte populations.

The HC10 monoclonal antibody (anti-human HLA Class I Heavy Chain) detected free HLA class I heavy chains on circulating monocytes via flow cytometry in patients with psoriatic arthropathy. Peripheral blood mononuclear cells, isolated by Ficoll-Hypaque centrifugation, were stained with HC10 followed by FITC-conjugated goat anti-mouse IgG and analyzed using a FACScan flow cytometer with CellQuest software.
