Description

The mouse IgG2c antibody is produced in certain inbred strains such as C57BL/6, C57BL/10, SJL, and NOD, which does not express the mouse IgG2a antibody, and vice versa. If one uses any of the mouse strains for animal research, it is better to use the IgG2c antibodies rather than the IgG2a antibodies. There are more and more IgG antibody formats with Fc engineered. The format with the Fc silenced, Fc silent, or Fcs with silenced effector function is the most popular. Thus, the wild type mouse IgG2c isotype control and the Fc silenced mouse IgG2c isotype control mutants should be used in such an application.

Background

Mouse IgG antibodies are classified to four subclasses mainly based on the isotype of the heavy chain (IgG1, IgG2a or IgG2c, IgG2b, and IgG3 for the heavy chain; kappa and lamda for the light chain), of which IgG1 and kappa are the most abundantnamed in serum. IgG antibodies are large molecules of about 150 kDa composed of four peptide chains, two identical heavy chains of about 50 kDa and two identical light chains of about 25 kDa. The four peptides are arranged in a Y-shape tetramer by disulfide bonds formed between the two heavy chains and between a heavy chain and a light chain.

Mouse IgG2c isotype control antibodies were used to ensure specificity of the observed effects in syngeneic tumor models. The mIgG2a isotype exhibited superior tumor growth inhibition compared to mIgG1 and mIgE in the MC38 model. All antibodies were administered intraperitoneally at a dose of 10 mg/kg twice weekly. Tumor volumes were measured using calipers every 3 days to assess treatment efficacy. The study highlights the importance of isotype selection in optimizing therapeutic antibody performance.

Design and Characterization of Mouse IgG1 and IgG2a Bispecific Antibodies for Use in Syngeneic Models.

A mouse IgG2c isotype control was included to validate the specificity of the bispecific antibody binding in vivo. The bispecific antibodies were designed to engage both CD3 and tumor-associated antigens in syngeneic mouse models. Binding affinity was determined using surface plasmon resonance with a Biacore T200 system. The IgG2a format showed enhanced effector function compared to IgG1 in T-cell activation assays. Pharmacokinetic studies confirmed the stability of the bispecific constructs in serum over 7 days.

Mouse IgG2a Antibodies Specific for the Commensal Streptococcus mitis Show Stronger Cross-Reactivity with Streptococcus pneumoniae than IgG1 Antibodies.

Mouse IgG2c isotype control antibodies were used to confirm the specificity of the immune response to Streptococcus mitis. IgG2a antibodies displayed stronger cross-reactivity with S. pneumoniae antigens compared to IgG1. Serum samples were collected from immunized mice at day 14 post-vaccination. ELISA assays were performed to quantify antibody titers against bacterial surface proteins. The study suggests that IgG2a antibodies may contribute to cross-protective immunity.

A mouse IgG2c isotype control was used to assess non-specific binding in flow cytometry experiments. Human IgE+ plasma cells were compared to IgG1+ counterparts using RNA sequencing. The transcriptional profile revealed distinct gene expression patterns in IgE+ cells. Cells were stained with fluorochrome-conjugated antibodies and analyzed on a BD LSRFortessa. The study provides insights into the molecular mechanisms of IgE-mediated allergic responses.

A Rapid Method to Characterize Mouse IgG Antibodies and Isolate Native Antigen Binding IgG B Cell Hybridomas.

Mouse IgG2c isotype-switched variants were generated to evaluate antigen-specific binding in hybridoma screening. The method allows for rapid isolation of native IgG-producing B cell hybridomas. Hybridomas were cultured in RPMI-1640 medium supplemented with 10% fetal bovine serum. Antigen-specific binding was confirmed using ELISA and flow cytometry assays. This approach significantly reduces the time required for antibody characterization.
