BP4091: In Vivo Grade Recombinant Human IgG1 Fc protein (hIgG1 Fc)

The in vivo grade recombinant human IgG1 Fc protein (hIgG1 Fc, Glu 99 - Lys 330 of the constant region of the IgG1 heavy chain, Accession # P01857-1) was produced in mammalian cells and purified by protein A. The hIgG1 Fc protein includes the CH2 (111-223) and CH3 (224-330) domains of the heavy chain constant region as well as the hinge region (99-110) before the CH2 domain. The C103S version of the hIgG1 Fc protein is also available. The C103S substitution prevents improper cysteine bond formation in the absence of a light chain.
Molecular weight: Predicted 26.1 kDa. Due to glycosylation, the recombinant human IgG1 Fc protein migrates to 32-34 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

Background

The recombinant human IgG1 Fc protein (hIgG1 Fc) includes the CH2 and CH3 domains of the heavy chain constant region. Most of the therapeutic antibody drugs and Fc fusion proteins carry the natural human IgG1 Fc region or engineered mutants. The recombinant human IgG1 Fc protein can be used as the isotype control of the hIgG1 antibodies and the hIgG1 Fc fusion proteins in various in vitro and in vivo experiments.

Changes to the IgG Fc sequence including glycosylation sites have been used to modulate Fc functions. Even a single mutation may have both enhancement and abrogation effects of Fc binding. The following mutations have been introduced to alter the Fc region of an IgG1 antibody or an IgG1 Fc fusion protein to change its immunologic properties by Fc interaction with host proteins:

PMN-MDSC were sorted as CD11b+ HLA-DR− CD15+ using FACS Aria II (BD Bioscience, USA), with Fc receptor blocking solution (BioLegend, USA) added prior to staining. Compared to M-MDSC, more PMN-MDSC accumulated in the decidua in both normal pregnancy (NP) and unexplained recurrent pregnancy loss (URPL) groups (P < 0.0001). Notably, decidual PMN-MDSC significantly decreased in the URPL group compared to normal pregnancy (P = 0.001). A significant difference was observed in the proportion of apoptotic cells in freshly isolated decidual PMN-MDSC between the NP and URPL groups (P = 0.004).

Engineering the Fc Region of Immunoglobulin G.

The human IgG1 Fc region mediates effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Mutations in the lower hinge region of human IgG1 Fc enhance ADCC by improving interaction with FcγRIIIa. The wild-type human IgG1 Fc protein was expressed in CHO cells and purified using protein A chromatography. Binding affinities to Fcγ receptors were measured via surface plasmon resonance, with engineered variants showing up to 10-fold increased affinity for FcγRIIIa.

Production of Human IgG1 Fc Fusion Proteins in CHO Cells.

Human IgG1 Fc fusion proteins were produced by transfecting CHO-DG44 cells with expression vectors containing the fusion gene. The human IgG1 Fc protein was fused to the C-terminus of the target protein to enable dimerization and extend half-life. Cells were cultured in serum-free medium and harvested after 7 days. Protein A affinity chromatography yielded >95% purity. The fusion proteins retained the biological activity of the target domain.

Structure-Function Analysis of Human IgG1 Fc Glycosylation.

Glycosylation at Asn297 in the human IgG1 Fc region is critical for effector functions. Deglycosylated human IgG1 Fc protein exhibited reduced binding to FcγRIIIa. The asialo-agalacto form of human IgG1 Fc showed anti-inflammatory properties in vivo, while sialylated forms were prepared by enzymatic remodeling. These glycoforms were tested in a mouse model of arthritis to evaluate their functional properties.

Engineering the human IgG1 Fc can modulate antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). Recombinant human IgG1 Fc protein was used to map binding epitopes, with mutations at Leu234 and Leu235 reducing effector functions. Modified human IgG1 Fc proteins were incorporated into monoclonal antibodies for clinical trials.
