In vivo grade recombinant natural and engineered mouse IgG3 Fc proteins (mIgG3 Fc), and recombinant mouse IgG3 isotype control antibody and mutants are available. Biotinylated mIgG3 Fc, and mIgG3 Fc with Avi-, His-, and DYKDDDDK-tags are also available. The recombinant mouse IgG3 Fc protein and mutants can be used as the isotype control of the mIgG3 antibodies and the mIgG3 Fc fusion proteins in various in vitro and in vivo experiments.

BP4124: In Vivo Grade Recombinant Mouse IgG3 Fc protein (mIgG3 Fc)

The in vivo grade recombinant mouse IgG3 Fc protein (mIgG3 Fc, Glu 97 - Lys 329 of the constant region of the IgG3 heavy chain, Accession # P03987-2) was produced in mammalian cells and purified by protein A. The mIgG3 Fc protein includes the CH2 (114-223) and CH3 (224-327) domains of the heavy chain constant region as well as the hinge region (97-113) before the CH2 domain.
Molecular weight: Predicted 26.2 kDa. Due to glycosylation, the recombinant mouse IgG3 Fc protein migrates to 32-35 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

No binding of mouse IgG1 to FcγRI and FcγRIV, no binding of mouse IgG2b to FcγRIII, and no binding of mouse IgG3 to any mouse FcγR was detected. Dissociation constants for mouse IgG subclasses at R max = 500 are shown in Figure 2. Anti-Kell mouse IgG subclasses were generated using the PUMA1 variable domain specific for Kell antigen. IgG was isolated from cell supernatant using HiTrap Protein A HP for human IgG1, IgG2, and IgG4, or HiTrap Protein G HP for human IgG3 and mouse IgG1, IgG2a, IgG2b, and IgG3 on ÄKTA prime. No binding of mouse IgG2b or IgG3 to mFcγRI was observed, despite some reported positive interactions.

The study confirmed that J774A.1 cells express a receptor binding antigen-free mouse IgG3, as reported by Saylor et al. Deglycosylation of mouse IgG3 prevented its binding to J774A.1 and P388D1 cells. Accutase treatment of cells also affected mouse IgG3 binding. Blockade of ITGB1 reduced mouse IgG3 binding to these cells. The study showed that mouse IgG3 binding depends on N-glycans in the CH2 domain of the antibody.

Mouse IgG1, IgG2a, IgG2b, and IgG3 specific for the C-terminal 19-kDa region of Plasmodium falciparum and Plasmodium yoelii MSP119 were generated to study protective mechanisms. These antibodies bound antigen with affinity comparable to their parental monoclonal antibodies. Mouse IgG1 recognizing PfMSP119 was effective in inducing NADPH-mediated oxidative bursts and neutrophil degranulation in vitro but failed to protect against parasite challenge in vivo in the Plasmodium berghei transgenic model, suggesting a minor role in infection control.

Mouse IgG3 antibodies uniquely form oligomers upon antigen binding through their Fc fragment. Divalent F(ab′)2 from pepsin-digested M18 retained agglutination capability, indicating that M18-induced haemagglutination is Fc-independent. IgG3s with M18 or O10 variable regions retained RBC agglutination, unlike corresponding IgG1 molecules. In silico analysis suggested that IgG3 Fabs may extend up to 3.4 nm longer than other isotypes. Mouse IgG3s are proposed as feasible alternatives to IgMs for blood group typing.

FcγRI is a high-affinity receptor for mouse IgG2a and a low-affinity receptor for mouse IgG2b and IgG3. All activating mouse FcγRs (FcγRI, FcγRIII, FcγRIV) bind IgG2a and IgG2b, while FcγRIII binds IgG1 and FcγRI binds IgG3. FcγRI contributes to IgG2a-induced type I and II hypersensitivity, collagen-induced arthritis, and reversed passive Arthus reaction. FcγRIII is critical for IgG1-induced passive systemic anaphylaxis and K/BxN arthritis. Mice expressing human FcγRs without endogenous FcγRs were used to study receptor properties without interference.
