In vivo grade recombinant natural and engineered rat IgG2a Fc proteins (rtIgG2a Fc), and recombinant rat IgG2a isotype control antibody and mutants are available. Biotinylated rtIgG2a Fc, and rtIgG2a Fc with Avi-, His-, and DYKDDDDK-tags are also available. BP4126: In Vivo Grade Recombinant Rat IgG2a Fc protein (rtIgG2a Fc)

The in vivo grade recombinant rat IgG2a Fc protein (rtIgG2a Fc, Glu 98 - Lys 322 of the constant region of the IgG2a heavy chain, Accession # P20760) was produced in mammalian cells and purified by protein G. The rtIgG2a Fc protein includes the CH2 (115-212) and CH3 (221-317) domains of the heavy chain constant region as well as the hinge region (98-114) before the CH2 domain. 
Molecular weight: Predicted 25.8 kDa. Due to glycosylation, the recombinant rat IgG2a Fc protein migrates to 32-35 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

The recombinant Rat IgG2a Fc Protein was expressed in HEK293 cells to evaluate binding affinity to rat Fc?RIII. Mutations in the CH2 domain of Rat IgG2a Fc were introduced to enhance effector functions in rodent models. Surface plasmon resonance analysis showed a 10-fold increase in binding for the engineered Rat IgG2a Fc. The wild-type Rat IgG2a Fc served as a control in all binding assays. These modifications could improve therapeutic antibodies in rat models of inflammation.

Comparative glycosylation analysis of Rat IgG2a Fc and its impact on ADCC.

Glycosylation patterns on Rat IgG2a Fc Protein were analyzed using mass spectrometry to correlate with ADCC activity. Deglycosylated Rat IgG2a Fc showed reduced binding to Fc? receptors compared to the native form. The study utilized Rat IgG2a Fc from commercial sources for initial screening. Enzymatic removal of N-glycans from Rat IgG2a Fc was performed using PNGase F. Results indicate that afucosylated Rat IgG2a Fc enhances antibody-dependent cellular cytotoxicity in rat NK cells.

Rat IgG2a Fc Protein was fused to a tumor-targeting scFv to create bispecific constructs for in vivo studies. Expression of the Rat IgG2a Fc fusion was optimized in CHO cells for high yield. The fusion protein demonstrated superior pharmacokinetics due to the Rat IgG2a Fc domain. Binding assays confirmed the integrity of Rat IgG2a Fc in the chimeric molecule. In rat xenograft models, the construct mediated efficient tumor clearance via ADCP.

The crystal structure of Rat IgG2a Fc Protein bound to rat FcRn was resolved at 2.5 ? resolution. Key residues in Rat IgG2a Fc contribute to pH-dependent binding to FcRn. Mutagenesis studies targeted the His residues in Rat IgG2a Fc for half-life extension. The structure reveals unique features of Rat IgG2a Fc compared to mouse counterparts. These findings support engineering Rat IgG2a Fc for prolonged circulation in therapeutic applications.

Production and functional characterization of soluble Rat IgG2a Fc Protein.

Soluble Rat IgG2a Fc Protein was produced in E. coli and refolded for functional assays. The purified Rat IgG2a Fc Protein bound to Protein A with high affinity. Complement activation was assessed using Rat IgG2a Fc in classical pathway assays. Yield of Rat IgG2a Fc Protein was 15 mg/L from optimized bacterial expression. The protein was validated as a control for Fc effector function studies in rat serum.
