Background

Human IgG4 Fc is the tail region of the immunoglobulin G4 (IgG4) that interacts with cell surface receptors called Fc receptors and some proteins of the complement system. The ~230 amino acid fragment generally exists as a dimer, although under reducing conditions, it exists as a monomer. It is the basis of prolonged pharmacokinetics of antibodies and is commonly used as a fusion to extend half-life of fusion proteins. IgG4’s hinge is shorter than that of IgG1, its flexibility is intermediate between IgG1 and IgG2 and some rotation may occur around the glycine residue in its hinge region.

The human IgG4 Fc protein was expressed in HEK293 cells and purified using protein A affinity chromatography to achieve high yield and purity. Glycoengineering at Asn297 improved binding to FcγRIIIa without altering FcRn affinity. In vitro ADCC assays demonstrated enhanced cytotoxicity with the modified human IgG4 Fc protein compared to wild-type. The engineered variant showed no aggregation issues during stability testing at 37°C. These modifications have potential applications in next-generation monoclonal antibodies.

Engineering Human IgG4 Fc for Reduced Inflammation in Autoimmune Diseases.

The human IgG4 Fc region was mutated at positions 234 and 235 to ablate effector functions. Purified human IgG4 Fc protein was used to confirm loss of C1q binding via ELISA. In vivo studies in a collagen-induced arthritis model showed reduced joint inflammation with the silenced human IgG4 Fc protein. Surface plasmon resonance analysis quantified the binding kinetics of human IgG4 Fc protein to Fcγ receptors. The human IgG4 Fc protein construct was cloned into a mammalian expression vector for transient transfection.

Human IgG4 Fc fusion proteins were expressed in Sf9 insect cells using the baculovirus system for rapid production. The human IgG4 Fc protein domain was fused to a therapeutic peptide to enhance serum half-life. SEC-HPLC analysis confirmed the dimeric structure of the purified human IgG4 Fc protein. Bioactivity assays validated the retained function of the target moiety. Yield optimization led to 50 mg/L of functional human IgG4 Fc protein from bioreactor cultures.

Glycosylation site-specific mutants of human IgG4 Fc protein were generated by site-directed mutagenesis. NMR spectroscopy revealed conformational changes in the human IgG4 Fc protein upon sialylation. The human IgG4 Fc protein was deglycosylated using PNGase F for comparative binding studies. Sialic acid-capped human IgG4 Fc protein suppressed DC maturation in vitro.

The human IgG4 Fc protein was knobbed at the CH3 interface for heterodimerization in bispecific formats. Knob-into-hole human IgG4 Fc protein pairs assembled with >90% efficiency in co-transfection. FcγR cross-linking assays confirmed equivalent effector silencing in the human IgG4 Fc scaffold. The modular human IgG4 Fc protein design allowed facile incorporation of additional binding domains. Preclinical pharmacokinetics showed extended half-life for the human IgG4 Fc-based multispecific antibody.
