In vivo grade recombinant natural and engineered mouse IgG2b Fc proteins (mIgG2b Fc), and recombinant mouse IgG2b isotype control antibody and mutants are available. Biotinylated mIgG2b Fc, and mIgG2b Fc with Avi-, His-, and DYKDDDDK-tags are also available. Please contact us to ask for a quote for the engineered mouse IgG2b Fc proteins, biotinylated mIgG2b Fc, and mIgG2b Fc with Avi-, His-, and Flag-tags. The recombinant mouse IgG2b Fc protein and mutants can be used as the isotype control of the mIgG2b antibodies and the mIgG2b Fc fusion proteins in various in vitro and in vivo experiments.

BP4121: In Vivo Grade Recombinant Mouse IgG2b Fc protein (mIgG2b Fc)

The in vivo grade recombinant mouse IgG2b Fc protein (mIgG2b Fc, Glu 97 - Lys 335 of the constant region of the IgG2b heavy chain, Accession # P01867-2) was produced in mammalian cells and purified by protein A. The mIgG2b Fc protein includes the CH2 (126-225) and CH3 (234-330) domains of the heavy chain constant region as well as the hinge region (98-125) before the CH2 domain. 
Molecular weight: Predicted 26.7 kDa. Due to glycosylation, the recombinant mouse IgG2b Fc protein migrates to 33-35 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

Mouse IgG subclasses cross-react with human Fc gamma receptors (hFc?Rs), showing substantial functional conservation despite interspecies differences. Binding of human IgG to mouse Fc?Rs has been used to study effector functions in mice, but mouse IgG binding to human Fc?Rs can cause misinterpretation in applications like immunostaining. This study documents that deglycosylation eliminates binding only for mouse IgG2b, highlighting its unique interaction profile with hFc?Rs.

N-Linked glycosylation of the Fc domain of mouse IgG is a significant modulator of antibody functions and is subclass-specific. This study used multiple reaction monitoring to quantify subclass-specific peptides and Fc N-glycopeptides for IgG1*, IgG2b/2b*, IgG2a/2c, and IgG3 in mixed mouse serum, prepared by adding mouse IgG standard to C57BL/6 mouse serum. All peptides except IgG2aA were detected, aiding the study of how Fc glycosylation affects IgG subclass effector functions in various physiological or pathological states.

Mouse IgG2a and IgG2b exhibit the greatest protective and pathogenic properties among mouse IgG subclasses, partly due to their binding to Fc?RIV, a novel ? chain-dependent activating Fc receptor. Fc?RIV, expressed on myeloid lineage cells, binds IgG2a and IgG2b with intermediate affinity but shows no binding to IgG1 or IgG3. Blocking Fc?RIV binding to pathogenic anti-platelet antibodies protected mice from antibody-induced thrombocytopenia.

A simple and rapid method for isolating pure mouse IgG1, IgG2a, and IgG2b immunoglobulins in nearly 100% yield is described. Mouse serum was fractionated on protein A-Sepharose 4B, with IgM, IgA, and IgE recovered in the effluent at pH 8.0. IgG subclasses were retained at pH 8.0 and sequentially eluted at pH 6.0?7.0 (IgG1), pH 4.5?5.0 (IgG2a), and pH 3.5?4.0 (IgG2b) using buffers of decreasing pH, provided the column's binding capacity was not exceeded.
