In vivo grade recombinant natural and engineered rabbit IgG Fc proteins (rIgG Fc), and recombinant rabbit IgG isotype control antibody and mutants are available. Biotinylated rIgG Fc, and rIgG Fc with Avi-, His-, and DYKDDDDK-tags are also available. Please contact us to ask for a quote for the engineered rabbit IgG Fc proteins, biotinylated rIgG Fc, and rIgG Fc with Avi-, His-, and Flag-tags. The recombinant rabbit IgG Fc protein and mutants can be used as the isotype control of the rIgG antibodies and the rIgG Fc fusion proteins in various in vitro and in vivo experiments.

BP4122: In Vivo Grade Recombinant Rabbit IgG Fc protein (rIgG Fc)

The in vivo grade recombinant rabbit IgG Fc protein (rIgG Fc, Cys 105 - Lys 323 of the constant region of the IgG heavy chain, Accession # P01870-1) was produced in mammalian cells and purified by protein A. The rIgG Fc protein includes the CH2 (114-213) and CH3 (222-323) domains of the heavy chain constant region as well as the hinge region (105-113) before the CH2 domain. 
Molecular weight: Predicted 24.8 kDa. Due to glycosylation, the recombinant rabbit IgG Fc protein migrates to 28-32 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

The crystal structure of rabbit IgG-Fc was determined at 2.7 ? resolution. Rabbit IgG Fc protein was expressed in HEK293 cells and purified using protein A affinity chromatography. The asymmetric unit contained two rabbit IgG-Fc molecules with slightly different conformations. The hinge region in rabbit IgG-Fc shows higher flexibility compared to human IgG-Fc. Structural comparison revealed unique features in the CH2 domain of rabbit IgG-Fc.

Key residues in the lower hinge region were conserved between human and rabbit IgG Fc.

The structure of a human Fc?RI/Fc?RIIA complex with IgG1 Fc reveals a novel mechanism of cooperative binding

The complex formation was stabilized by interactions similar to those in rabbit IgG Fc. Structural overlays with rabbit IgG Fc highlighted conserved motifs. Mutagenesis studies used rabbit IgG Fc protein to confirm functional residues.

Crystal structure of rabbit IgG Fc and its implications for Fc engineering

Rabbit IgG Fc protein was crystallized in the orthorhombic space group P2(1)2(1)2(1). The structure reveals a unique conformation in the CH2-CH3 interface of rabbit IgG Fc protein. Engineering mutations were designed based on rabbit IgG Fc protein scaffold. Comparison with human IgG Fc showed differences in glycosylation sites. The resolved structure aids in Fc engineering for therapeutic applications.

Implications for antibody engineering are discussed based on this rabbit IgG Fc structure.
