In vivo grade recombinant natural and engineered rat IgG2b Fc proteins (rtIgG2b Fc), and recombinant rat IgG2b isotype control antibody and mutants are available. Biotinylated rtIgG2b Fc, and rtIgG2b Fc with Avi-, His-, and Flag-tags are also available. The recombinant rat IgG2b Fc protein and mutants can be used as the isotype control of the rtIgG2b antibodies and the rtIgG2b Fc fusion proteins in various in vitro and in vivo experiments.

BP4123: In Vivo Grade Recombinant Rat IgG2b Fc protein (rtIgG2b Fc)

The in vivo grade recombinant rat IgG2b Fc protein (rtIgG2b Fc) was produced in mammalian cells and purified by protein G. The rtIgG2b Fc protein includes the CH2 and CH3 domains of the heavy chain constant region as well as the hinge region before the CH2 domain. 
Molecular weight: Predicted 25.8 kDa. Due to glycosylation, the recombinant rat IgG2b Fc protein migrates to 32-35 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

Mouse IgG subclasses cross-react with human Fc gamma receptors (hFc?Rs). Deglycosylation of mouse IgG1 did not abrogate binding to hFc?RIIa-R131, nor did deglycosylation of mIgG2a/c and mIgG3 prevent hFc?RIa binding. However, deglycosylation of mouse IgG2b abrogated reactivity to all hFc?Rs. The study documents the full spectrum of cross-reactivities of mouse IgG to human Fc?Rs, noting that such cross-reactivities can cause misinterpretation in applications like immunostaining with mouse IgG monoclonal antibodies.

Rat IgG mediated circulatory cell depletion in mice requires mononuclear phagocyte system and is facilitated by complement.

Rat monoclonal antibodies, including anti-C5aR (20/70, IgG2b), anti-VLA4 (PS/2, IgG2b), anti-CD11b (M1/70, IgG2b), and anti-CD11a (M17/4, IgG2a), were tested for cell depletion effects in mice. The study demonstrates that Ab-mediated cell depletion, reliant on the Fc portion, is an efficient tool for studying cell function under normal and disease conditions. The mononuclear phagocyte system and complement facilitate depletion, with Rat IgG2b antibodies showing significant activity.

Mouse IgG1 and rat IgG2a Fcs preferentially bind to mouse Fc?RIIB, while mouse IgG2a and rat IgG2b Fcs bind to activating Fc?Rs. A non-depleting rat IgG2a Fc anti-mouse CD27 antibody was more effective than a depleting rat IgG2b Fc clone in antitumor responses against CD27+ and CD27- EL4 syngeneic tumors. The study supports a general requirement for Fc?RIIB co-engagement, rather than activating Fc?Rs, for agonistic anti-TNFR antibodies, highlighting the role of Fc interactions in therapeutic efficacy.

Bispecific antibodies (bsAbs) were generated by fusing mouse IgG2a and rat IgG2b hybridomas, exploiting species-restricted heavy-light chain pairing and differential Protein A elution to increase yield (mean recovery 75.2%). The study evaluated mutations in rat IgG1, IgG2a, IgG2b, and IgG2c backbones, confirming their applicability. This approach simplifies purification and enhances the production of bsAbs for pre-clinical studies in syngeneic rodent models.
