BP4119: In Vivo Grade Recombinant Llama IgG2b Fc protein (lIgG2b Fc)

The in vivo grade recombinant llama IgG2b Fc protein (lIgG2b Fc, Glu 1 - Lys 243 of the constant region of the IgG2a heavy chain, Accession # AAX73259.1) was produced in mammalian cells and purified by protein A. The lIgG2b Fc protein includes the CH2 (28-137) and CH3 (138-243) domains of the heavy chain constant region as well as the hinge region (1-27) before the CH2 domain. 
Molecular weight: Predicted 27.3 kDa. Due to glycosylation, the recombinant llama IgG2b Fc protein migrates to 35-40 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

For the RBD nanobody discovery campaign, 15 ?g RBD-llama IgG2b Fc fusion protein was administered subcutaneously in 200 ?l SAS-emulsion, and 15 ?g antigen in 150 ?l PBS containing 50 ?g poly I:C was injected intraperitoneally. Mice received five boost immunizations at 2?4-week intervals. By inserting an engineered llama immunoglobulin heavy chain (IgH) locus into IgH-deficient mice, a transgenic mouse line, ‘LamaMouse,’ was generated to produce nanobodies with exceptional solubility and stability, offering a cost-effective alternative to llama-based nanobody production.

Monomeric or trimeric nanobody sequences were fused to the Fc region of human IgG1 with a 6×His tag at the C-terminal end, or in some cases, a llama IgG2a hinge region was used instead, and cloned into the pVRC8400 vector. Trimeric nanobodies were connected via (GGGGS)×3 flexible linkers. Proteins were filtered through a 0.22-?m PVDF membrane before assays. For larger-scale production (0.2 to 1 L), nanobodies were purified using cOmplete His-tag purification resin and dialyzed in PBS.

A variable fragment of a heavy chain antibody (VHH), anti-rotavirus protein 1 (ARP1), was fused to the mouse IgG1 Fc to enhance protection against rotavirus-induced diarrhea in an infant mouse model. Co-expression of anti-rotavirus VHH fragments in Lactobacillus was developed with two expression cassettes in tandem. This fusion improved the protective capacity of ARP1 by inducing Fc-mediated effector functions, highlighting the potential of antibody-based drugs as alternatives to standard antifungals with high toxicity and failure rates.

Polyclonal alpaca antibodies protect against hantavirus pulmonary syndrome in a lethal Syrian hamster model.

Polyclonal alpaca IgG antibodies (Alp-PcIgG-ANDV), including IgG1 and IgG3, were purified using the AKTA Pure Protein Purification system with a HiTrap Protein A column. The glycoprotein precursor coding region of ANDV Chile 9717869 was cloned into the pCAGGS plasmid, and protein expression was confirmed by Western blot using monoclonal antibodies targeting ANDV glycoproteins. The study suggests that increased DNA concentrations or additional time points may not significantly affect specific antibody yields.

The pharmacokinetic properties of antibodies are influenced by pH-dependent binding of the IgG Fc domain to the human neonatal Fc receptor (hFcRn). An engineered IgG1 Fc domain with enhanced hFcRn binding was developed, acting as a pH-toggle switch to extend circulation half-life. Biophysical studies elucidated the role of CH2?CH3 interface residues, His310, and His435 in pH-dependent binding. The engineered Fc domain improved pharmacokinetics in humanized knock-in mouse strains, relevant for antibody persistence studies.
