Description

The recombinant human IgG2 isotype control (hIgG2 kappa isotype control antibody) is produced in mammalian cells (HEK293 or CHO cells) like our other recombinant human IgG isotype control and mutant antibodies. The ideal recombinant human IgG2 isotype control has low or no specific binding to any human sample, and humanized or completely human variable regions in addition to the human IgG2 kappa constant regions. The human IgG2 kappa isotype control can be used in the cell-based assay or animal model as the antibody negative control.

Background

Naturally synthesized and secreted by plasma B cells, the immunoglobulin G (IgG) antibody is the most abundant antibody isotype found in blood and extracellular fluid. There are four IgG subclasses (IgG1, IgG2, IgG3, and IgG4) in humans, of which IgG1 is the most abundantnamed in serum. IgG antibodies are large molecules of about 150 kDa composed of four peptide chains, two identical heavy chains of about 50 kDa and two identical light chains of about 25 kDa. The four peptides are arranged in a Y-shape tetramer by disulfide bonds formed between the two heavy chains and between a heavy chain and a light chain.

The arms of the Y, also called the Fab (fragment, antigen-binding) region containing an identical antigen binding site, is composed of one variable (located at amino terminal end, VH and VL) and one constant domain (CH1 and CL) from each heavy and light chain of the antibody. VH and VL, also called the FV region, is the most important region for binding to antigens. Three variable loops of β-strands, also called the complementarity determining regions (CDRs or idiotypes), on VH and VL respecetively are responsible for binding to the antigen.

The base of the Y, also called the Fc (Fragment, crystallizable) region and composed of two heavy chains with two or three constant domains depending on the class of the antibody, binds to a specific class of Fc receptors and other immune molecules, such as complement proteins, to induce an appropriate immune response for a given antigen. The Fc region bears two highly conserved N-glycosylation sites, one on each heavy chain. The N-glycans attached to this site are predominantly core-fucosylated diantennary structures of the complex type. In addition, small amounts of these N-glycans also bear bisecting GlcNAc and α-2,6-linked sialic acid residues.

Immunoglobulin G (IgG) antibodies, the most abundant isotype in healthy human serum, are divided into four subclasses: IgG1 (60–70% in plasma), IgG2 (20–30%), IgG3 (5–8%), and IgG4 (1–3%). IgG antibodies are critical for neutralizing pathogens and other foreign substances in the adaptive immune system. Structural differences, such as disulfide bond linkages between heavy and light chains, distinguish the subclasses. Currently, only IgG1, IgG2, and IgG4 are considered for therapeutic purposes, but IgG3, the most potent in mediating Fc effector functions, and IgG polymorphisms could enhance antibody-mediated effector functions through differential binding to FcγRs and complement proteins. An example is the afucosylated humanized IgG1 antibody benralizumab (MEDI-563), which targets IL-5 Receptor α-chain for severe eosinophilic asthma.

Human IgG2 antibody exhibits distinct therapeutic properties compared to IgG1, IgG3, and IgG4 subclasses. As the second most abundant IgG subclass, IgG2 binds to human FcγRII/FcγRIII but not to FcγRI or complement C1q. The solution structure of human myeloma IgG2 was determined using atomistic X-ray and neutron-scattering modeling. Analytical ultracentrifugation showed that IgG2 is monomeric with a sedimentation coefficient (s20, w0) of 7.2 S, with dimer formation ≤5% and independent of buffer conditions.

In C57BL/6 mice, evaluating IgG2c antibody alongside IFN-γ is critical for accurate interpretation of Th1 immune responses. Measuring murine isotype antibodies is essential in subunit vaccine development due to varied responses to recombinant proteins across inbred mouse strains with diverse genetic backgrounds. IFN-γ enhances antibody production, correlating with increased parasite-specific IgG2a levels. Since mouse IgG heavy chain constant regions are encoded by tightly linked genes inherited as Igh haplotypes, this study measured and compared IgG2a and IgG2c isotypes, their avidities, titration, and cytokines induced by recombinant PvTRAP antigen in C57BL/6 mice to clarify Th1/Th2 immune responses.

Whole-body immunoPET reveals active SIV dynamics in viremic and antiretroviral therapy–treated macaques.

ImmunoPET/CT imaging using a labeled SIV-specific antibody identified sites of viral infection in SIV-infected macaques without requiring tissue biopsies, even when blood viral loads were below detection limits. A modified isotype control IgG antibody in a control macaque showed significantly lower background signal compared to infected animals, particularly in the liver, heart, and kidneys. Single-plane images and SUVmax quantification of modified mouse IgG in chronically infected and non-infected animals showed similar uptake, confirming the specificity of the SIV-specific antibody.

In macaques, CH31 IgG1 and IgA2 isoforms infused before high-dose SHIV challenge were completely to partially protective, respectively, while non-neutralizing antibodies (CH54 IgG1 and CH38 mIgA2) were non-protective. IgG1 isotype broadly neutralizing antibodies (bnAbs) were more protective than IgA2 isotypes in mucosal models, attributed to greater neutralization activity. The study used a human ex vivo vaginal HIV-1 infection model and a rhesus macaque in vivo intrarectal SHIV challenge model to assess the protective capacity of functionally modified monoclonal bnAbs and non-neutralizing antibodies through isotype switching.
