Background

Rat IgG antibodies are classified to four subclasses mainly based on the isotype of the heavy chain (IgG1, IgG2a, IgG2b, and IgG2c for the heavy chain; kappa and lamda for the light chain), of which IgG1 and kappa are the most abundantnamed in serum. IgG antibodies are large molecules of about 150 kDa composed of four peptide chains, two identical heavy chains of about 50 kDa and two identical light chains of about 25 kDa. The four peptides are arranged in a Y-shape tetramer by disulfide bonds formed between the two heavy chains and between a heavy chain and a light chain.

Rats have four IgG subclasses (IgG1, IgG2a, IgG2b, IgG2c), with unknown subclass-specific glycosylation variability. A cohort of 80 animals exposed to seven stressors showed increased agalactosylated glycoforms on IgG2a and IgG2c and decreased monogalactosylation in young female rats. A high-throughput glycoproteomic workflow for rat IgG Fc N-glycosylation analysis revealed N-glycosylation diversity in IgG2a, IgG2b, and IgG2c subclasses, with satisfactory repeatability and reproducibility.

Rat IgG mediated circulatory cell depletion in mice requires mononuclear phagocyte system and is facilitated by complement.

Mice (n = 5 mice/group) were intraperitoneally injected with 100 µg of rat anti-CD88 (C5aR), anti-VLA4, anti-CD11b, or anti-CD11a mAbs or isotype control antibodies. Ab-mediated complement activation relies on the Fc portion, enabling cell depletion regardless of the Fab portion. After 12 hours, vascular leukocyte depletion was analyzed by flow cytometry, comparing relative depletion to isotype control and antigen expression on cell types.

In C57BL/6 mice, evaluating IgG2c alongside IFN-γ is critical for accurate Th1 immune response interpretation. Mouse IgG2a and IgG2c are distinct isotypes encoded by tandem loci. The study measured IgG2a and IgG2c isotypes, their avidities, and cytokines induced by recombinant PvTRAP antigen. Immunized mice with rPvTRAP or rPvTRAP + CFA/IFA showed intermediate-avidity IgG2a but high-avidity IgG2c, with IgG2c end-point titers significantly higher than IgG2a.

Rat IgG2c proteins lack C1 binding and anticomplementary activity. IgM, IgG1, and IgG2a bind C1 and consume total complement, while IgA activates the alternative pathway without C1 binding. Staphylococcal protein A binds IgG1 and IgG2c but not IgG2a or IgG2b. The study tested monoclonal rat immunoglobulins and normal serum fractions for complement interaction and protein A binding.

Rat IgG subclasses: Differences in affinity to protein A-Sepharose.

The binding of rat IgG subclasses (IgG1, IgG2a, IgG2b, IgG2c) to protein A-Sepharose showed affinity differences: IgG2c > IgG1 > IgG2b > IgG2a, with IgG2a having no affinity and IgG2b having very weak affinity. Monoclonal IgG2c exhibited heterogeneity, with fractions eluted at pH 4.0–3.0 or pH 6.0. Normal rat serum IgG subclasses followed similar elution patterns, confirming the results from monoclonal IgG.
