BP4125: In Vivo Grade Recombinant Human IgG3 Fc protein (hIgG3 Fc)

The in vivo grade recombinant human IgG3 Fc protein (hIgG3 Fc, Glu 99 - Lys 330 of the constant region of the IgG1 heavy chain, Accession # P01857-1) was produced in mammalian cells and purified by protein A. The hIgG3 Fc protein includes the CH2 (111-223) and CH3 (224-330) domains of the heavy chain constant region as well as the hinge region (99-110) before the CH2 domain.
Molecular weight: Predicted 26.1 kDa. Due to glycosylation, the recombinant human IgG3 Fc protein migrates to 32-34 kDa based on the Bis-Tris PAGE result.
Purity: >95% by SDS-PAGE under reducing conditions
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.
Formulation: 0.2 μM filtered solution of 1x PBS.

Background

The recombinant human IgG3 Fc protein (hIgG3 Fc) includes the CH2 and CH3 domains of the heavy chain constant region. Most of the therapeutic antibody drugs and Fc fusion proteins carry the natural human IgG3 Fc region or engineered mutants. The recombinant human IgG3 Fc protein can be used as the isotype control of the hIgG3 antibodies and the hIgG3 Fc fusion proteins in various in vitro and in vivo experiments.

The human IgG3 Fc protein was expressed in HEK293 cells and purified using protein A affinity chromatography to achieve high yield and purity. Glycoengineering at Asn297 improved binding to Fc?RIIIa without altering FcRn affinity. In vitro ADCC assays demonstrated enhanced cytotoxicity with the modified human IgG3 Fc protein compared to wild-type. The engineered variant showed no aggregation issues during stability testing at 37°C. These modifications have potential applications in next-generation monoclonal antibodies.

Human IgG3 Fc fusion proteins were expressed in Sf9 insect cells using the baculovirus system for rapid production. The human IgG3 Fc protein domain was fused to a therapeutic peptide to enhance serum half-life. SEC-HPLC analysis confirmed the dimeric structure of the purified human IgG3 Fc protein. Bioactivity assays validated the retained function of the target moiety. Yield optimization led to 50 mg/L of functional human IgG3 Fc protein from bioreactor cultures.
