Source: The anti-mouse CD3e monoclonal antibody (clone: KT3) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: KT3) specifically binds to the mouse T cell receptor CD3e.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD3e protein.

CD1-restricted CD4+ T cells in major histocompatibility complex class II-deficient mice

Labeled cells were analyzed on FACScan | (Becton Dickinson & Co., Mountain View, CA), ODAM ATC 3000 (Bruker Spectrospin, Wissembourg, Ger- many), or Coulter Epic Elite (Coulter Corp., Hialeah, FL) flow cytometers, and sorted using the ATC or Elite. The sorted CD4 + cells were stimulated at 1.5-3 X 104 cells per well in anti- CD3-coated (KT3 antibody coated at 5 Ixg/ml or as indicated) round-bottom, 96-well plates in Hepes-buffered RPMI 1640 medium containing glutamine supplemented with 10% FCS, 20 IxM 2-ME, 1 m M Na-pyruvate, and penicillin/streptomycin. Intrathymic FITC injections were performed as described (19). Both thymic lobes of the 5-wk-old mice were injected with 10 txl of 1 mg/ml FITC in PBS. 30 h later, lymph nodes were analyzed for the presence of thymus-derived, F I T C - labeled cells (Fig. 3 A). C D 4 + and C D 8 + cells from control animals and C D 8 + cells from mutants were all labeled to a similar extent.

Anti-CD3 therapy permits regulatory T cells to surmount T cell receptor-specified peripheral niche constraints

For anti-CD3 treatment, mice were injected i.v. with 3 ?g/g BW of anti-CD3 mAb (clone; KT3; protein G purified) for 5 consecutive days. For T reg depletion in BR.Foxp3 DTR mice, 50 ?g/g BW of DT was administered at days 1, 2, 4, 6, and 8). For blocking studies, 75 ?g of anti?IL-2 (clone; JES6-1A12; BioLegend), anti?IL-7Ra (clone; A7.R34.2.2; protein G purified), and anti?IL-15 (clone; AIO.3; eBioscience) were injected ip for 6 consecutive days, starting on the same day as anti-CD3. 0.5 mg of anti-TGF ? (clone; 2G7; protein G purified) was administered every other day for 2 wk. In vivo BrdU incorporation, 0.01 mg/g BW of BrdU was injected i.p., 6 h before euthanasia.

Fc receptor binding of anti-CD3 monoclonal antibodies is not essential for immunosuppression in vivo

Interestingly, this was not observed with the anti-CD3 mAb 17A2 (rat IgG2b) [10] and KT3 (rat IgG2a) [6, 11]. Our previous study showed that the non-mitogenic rat anti-CD3 mAb 17A2 and KT3 are equally effective as the mitogenic 145-2C11 mAb in suppressing skin allograft rejection [6]. Administration of 10 pg hamster anti-CD3 mAb 145-2C11 in vivo induced a strong increase in serum TNF-a levels (Fig. 3). To determine the role of FcyR in this TNF-a release, we temporarily blocked FcyR by giving 250 pg 2.462 mAb 12 to 18 h before 145-2C11 injection. The day after 145-2C11 injection, C57BL/Ka mice received an MHC class II-disparate B6.C-H-2bm12 skin graft.

Tolerance induction in double specific T-cell receptor transgenic mice varies with antigen

We have now studied T-cell tolerance in transgenic mice expressing a T-cell receptor with double specificities for lymphocytic choriomeningitis virus (LCMV)-H-2D b and for the mixed-lymphocyte stimulatory (Mls a) antigen. By contrast, tolerance to Mls a antigen in the same ??TCR transgenic Mls a mice leads to deletion of only mature thymocytes and peripheral T cells and does not affect CD4 +CD8 + thymocytes. Thus the same transgenic TCR-expressing T cells may be tolerized at different stages of their maturation and at different locations in the thymus depending on the antigen involved. In vivo adjuvant-induced mobilization and maturation of gut dendritic cells after oral administration of cholera toxin

Spleen and MLN cells were stained with the following mAbs: FITC-anti-CD11c (HL3), PE-anti-CD8? (53-6.7), PE-anti-CD11b (M1/70), PE-anti-B220 (RA3-6B2), PE-anti-CD3 (KT3), biotin-anti-CD40 (FGK45.5), biotin-anti-CD80 (RMMP-1), biotin-anti-CD86 (RMMP-2), biotin-anti-DEC-205 (NLDC145), and biotin-anti-CCR7 (4B12). To analyze the activation state of DC in vivo, 24 h after CT administration, 5 × 10 5 PP, LP, or MLN DC were incubated with 5 × 10 5 BALB/c splenocytes for 4 days, and proliferation was assessed by [3H]TdR incorporation. To analyze the ability of DC to present OVA in vivo, mice were fed with 10 mg of OVA in 0.2 ml of NaHCO3 3%, with or without 10 ?g of CT. To analyze the migration of DC in vivo, mice were fed with 10 mg of FITC-OVA (Molecular Probes, Eugene, OR), in 0.2 ml of NaHCO3 3%, with or without 10 ?g of CT. The animals were maintained under specific pathogen-free conditions in isolators.
