Isotype: Mouse IgG1 lambda.
Source: The anti-human CD3 monoclonal antibody (clone: SP34-2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: SP34-2) specifically binds to the human T cell receptor CD3.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human CD3 protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.

Background

The anti-human CD3 antibody (clone: SP34-2), a derivative of SP34, is known to cross-react with a major subset of peripheral blood lymphocytes in both human and non-human primates, including baboon, rhesus, cynomolgus, and pigtail macaque monkeys. The SP34-2 antibody specifically recognizes a major subset of CD3-positive cells from non-human primates, similar to its human counterpart, but does not react with monocytes or granulocytes. 

The SP34-2 antibody is used in flow cytometry to identify and analyze CD3-positive cells in human and various non-human primate species. It can also be used to stimulate cell proliferation in various primate peripheral blood mononuclear cells (PBMC).

This study investigates the role of Nef in Simian Immunodeficiency Virus (SIVagm) in African green monkeys (AGMs), which exhibit low immune activation despite high viremia. Flow cytometry was used to analyze cells stained with LIVE/DEAD Fixable Aqua, anti-human CD3 (SP34-2), CD4 (L200), CD8 (SK1), and CD20 (L27) antibodies. Virus stocks were generated by transfecting 293T cells with plasmid DNA encoding full-length SIVagm proviruses. Peripheral blood mononuclear cells (PBMCs) were isolated from whole blood using Ficoll gradient centrifugation, and viral RNA was extracted from plasma using the QIAamp Viral RNA minikit.

Autologous humanized mouse models of iPSC-derived tumors enable selection of immunotherapy candidates

This study explores autologous humanized mouse models generated from human induced pluripotent stem cells (iPSCs) to evaluate immunotherapy candidates. Single-cell suspensions were stained with LIVE/DEAD Fixable Aqua Dead Cell Stain Kit and anti-human antibodies: CD45 (HI30), CD3 (SP34-2), CD4 (OKT4), CD8 (SK), and PD-1 (EH12.2H7). Human iPSCs were derived from peripheral blood mononuclear cells (PBMCs) using Sendai virus-based reprogramming. Tumor growth was monitored via caliper measurements and bioluminescence imaging, with immune cell infiltration analyzed by flow cytometry in tumor, spleen, and bone marrow samples.

This study describes the development and preliminary clinical activity of MGD019, a bispecific molecule targeting PD-1 and CTLA-4. Flow cytometry was conducted using anti-CD3 (SP34-2), anti-CD4 (L200), and anti-CD8 (RPA-T8) antibodies. Cynomolgus monkeys received MGD019 at doses of 10 to 100 mg/kg weekly for 4 weeks. Peripheral blood samples were collected for pharmacokinetic analysis, and immune cell populations were quantified in spleen and lymph nodes at necropsy. Binding to PD-1 and CTLA-4 was confirmed using surface plasmon resonance.

Modified Vaccinia Virus Ankara Encoding Influenza Virus Hemagglutinin Induces Heterosubtypic Immunity in Mice

This study evaluates a Modified Vaccinia Virus Ankara (MVA) vaccine encoding influenza virus hemagglutinin in mice. Splenocytes were stained with anti-CD3 (SP34-2), anti-CD4 (L200), and anti-CD8 (SK1) antibodies for flow cytometric analysis. Mice were vaccinated intramuscularly with 10^7 PFU of MVA-HA or control MVA, followed by an influenza virus challenge with 10^5 TCID50 of H1N1 three weeks later. Lung viral titers were measured by plaque assay, and IFN-? production by T cells was assessed via ELISPOT assay.

CD8+ cellular immunity mediates rAd5 vaccine protection against Ebola virus infection of nonhuman primates

This study examines the role of CD8+ cellular immunity in rAd5 vaccine protection against Ebola virus in cynomolgus macaques. Lymphocytes were stained with anti-CD3 (SP34-2), anti-CD8 (SK1), and anti-IFN-? (B27) antibodies for intracellular cytokine staining. Macaques were vaccinated with rAd5 expressing Ebola virus glycoprotein and challenged with 1000 PFU of Ebola virus 4 weeks later. Peripheral blood was collected weekly for immune response monitoring via flow cytometry, with survival tracked daily and necropsy performed on nonsurvivors.
