Background:
The 12F6 antibody competes with the OKT3 antibody for binding to human T cells, and has more effective T-cell suppression and activation properties than OKT3. The 12F6 antibody is used to study T-cell functions and to manipulate T-cell activation and suppression, prevent acute allograft rejection, and treat autoimmune diabetes.
Polymorphic residues in HLA-B that mediate HIV control are associated with enhanced structural rigidity

T2 cells pulsed with 100 μM peptide were stained with anti-human CD3 (clone 12F6) and analyzed by flow cytometry to confirm T cell activation. Peptide binding to HLA-B was quantified using a fluorescence-based assay to assess structural rigidity. The study identified specific HLA-B residues that enhance peptide-MHC stability in HIV controllers. Flow cytometry data showed increased CD3 expression in T cells from HIV controllers compared to progressors. Structural analysis revealed that polymorphic residues contribute to a rigid HLA-B conformation, impacting T cell responses.

Simultaneous assessment of cytotoxic T lymphocyte responses in individuals with chronic hepatitis B virus infection

PBMCs were stained with anti-CD3 (clone 12F6) antibody and anti-CD8 (clone RPA-T8) for flow cytometric analysis of CTL responses. HBV-specific CTL responses were measured using peptide-MHC tetramers in chronic HBV patients. The frequency of CD8+ T cells producing IFN-γ was significantly higher in patients with controlled HBV infection. Flow cytometry revealed distinct T cell subsets in responders versus non-responders, highlighting the role of cytotoxic T lymphocytes in HBV immune control.

Rapid, efficient functional characterization and recovery of HIV-specific human CD8+ T cells using microengraving

After microengraving, cells were stained with fluorescently labeled antibodies against CD3 (clone 12F6, anti-human CD3e antibody) and CD8 to identify HIV-specific T cells. Microengraving allowed rapid identification and recovery of antigen-specific T cells. HIV-specific CD8+ T cells were sorted based on cytokine secretion profiles. The technique enabled high-throughput analysis of T cell functionality, with flow cytometry confirming the specificity of recovered T cells to HIV epitopes.

Phase I trial of a humanized, Fc receptor nonbinding anti-CD3 antibody, visilizumab (Nuvion) in treatment-resistant severe ulcerative colitis

Flow cytometry analysis used anti-CD3 (clone 12F6) antibody to monitor T cell depletion in peripheral blood samples from ulcerative colitis patients. Visilizumab treatment resulted in significant reduction of circulating T cells within 24 hours. The study evaluated the safety and efficacy of visilizumab in severe ulcerative colitis, noting cytokine release syndrome in a subset of patients. T cell recovery was monitored over 28 days using flow cytometry.
