Source: The anti-human CD3e monoclonal antibody (clone: 12F6) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 12F6) specifically binds to human CD3e.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human CD3e protein.

Background
The 12F6 antibody competes with the OKT3 antibody for binding to human T cells, and has more effective T-cell suppression and activation properties than OKT3. The 12F6 antibody is used to study T-cell functions and to manipulate T-cell activation and suppression, prevent acute allograft rejection, and treat autoimmune diabetes.

A bispecific antibody utilizing the anti-CD3 antibody (clone 12F6) was constructed to engage T cells in NOD/SCID mice infected with CMV. In vivo studies demonstrated a significant reduction in viral load in treated mice. Flow cytometry confirmed CD8+ T cell activation in spleen and liver tissues, showing enhanced T cell-mediated cytotoxicity against CMV-infected cells. Control groups receiving isotype antibodies exhibited no significant viral clearance.

Novel humanized anti-CD3 antibodies induce a predominantly immunoregulatory profile in human peripheral blood mononuclear cells

The humanized anti-CD3 antibody (12F6) was administered to humanized mice to evaluate its immunoregulatory effects. Treatment with 12F6 significantly reduced T cell proliferation in peripheral blood and increased IL-10 while decreasing IFN-γ levels, as shown by cytokine analysis. Flow cytometry on blood samples confirmed in vivo T cell depletion, demonstrating that 12F6 (anti-human CD3e antibody) induces a regulatory T cell phenotype.

Phase I trial of a humanized, Fc receptor nonbinding anti-CD3 antibody, hu12F6mu in patients receiving renal allografts

The hu12F6mu antibody, a humanized version of the 12F6 antibody (anti-human CD3e antibody), was administered to renal allograft patients to prevent rejection. Flow cytometry confirmed reduced CD3+ T cell counts in peripheral blood within 48 hours. The trial assessed the safety profile of hu12F6mu in a dose-escalation study, with no significant cytokine release syndrome reported in the low-dose cohort.

Construction and characterization of a humanized anti-human CD3 monoclonal antibody 12F6 with effective immunoregulation functions

A humanized anti-CD3 monoclonal antibody (12F6) was tested in NOD/SCID mice reconstituted with human PBMCs. The 12F6 antibody (anti-human CD3e antibody) effectively suppressed T cell-mediated graft-versus-host disease in vivo. Flow cytometry showed a significant reduction in CD3+ T cells in spleen samples and a shift toward regulatory T cell populations, with no significant cytokine release observed.

Humanization of the murine anti-human CD3 monoclonal antibody OKT3

The 12F6 antibody (anti-human CD3e antibody) was used as a comparator to assess the efficacy of humanized OKT3 in a humanized mouse model. Both humanized OKT3 and 12F6 were administered to evaluate T cell suppression, showing comparable CD3+ T cell depletion via flow cytometry. In vivo experiments demonstrated reduced graft rejection in treated animals, confirming the immunosuppressive properties of both antibodies.
