This study investigates the use of monovalent anti-CD3 antibodies, OKT3 and UCHT1, to eliminate TCR-positive allogeneic CAR-T cells to prevent graft-versus-host disease (GVHD). Full-length OKT3-IgG and UCHT1-IgG were purchased from Bio X Cell and BioLegend, respectively. Fab fragments were generated by cloning VH-CH1 and VL-CL domains into pCEP4 plasmids and transfecting FreeStyle? 293-F cells. Single-chain variable fragments (scFvs) for OKT3, UCHT1, and other clones were cloned into a pCEP4 vector with a human kappa chain constant region. TCR deletion was assessed by flow cytometry using anti-TCR??-PE/Cy5 (IP26) and anti-CD3?-PE (UCHT1) antibodies. Unlike full-length UCHT1-IgG, UCHT1-scFv induced significant apoptosis in expanded T cells.

This study employs full-spectrum flow cytometry to analyze human monocyte subsets, utilizing a panel of monoclonal antibodies including PerCP-conjugated CD45, BV510-conjugated CD3, and others detailed in the study. T cells were isolated using a CD3/CD14 plot, with CD3+ T cells further divided into ??/?? T cells via a CD3/TCR-?? plot. NKT cells were distinguished from ?? T cells using a CD56/CD3 plot. CD19+ B cells were defined within the CD3+ T cell-excluded PBMC compartment. The study provides a comprehensive analysis of immune cell populations using hierarchical marker clustering.

This study describes a flow cytometry assay to detect recipient IgG subtype antibodies binding to donor cells, improving donor availability for highly sensitized patients. The assay uses a lyophilized reagent containing PE/Cy7-conjugated anti-human CD3 antibody (clone UCHT1) for T cells and APC/Fire? 750-conjugated anti-human CD19 antibody (clone HIB19) for B cells. Cells were stained with a six-color lyophilized antibody reagent and analyzed on a BD FACSLyric? system using BD FACSuite? RUO v1.4 software. A separate compensation control for T cells was prepared with PE/Cy7-conjugated anti-CD3 antibody.

A flow cytometry-based proliferation assay for clinical evaluation of T-cell memory against SARS-CoV-2

This study presents a flow cytometry-based proliferation assay (FASCIA) to evaluate T-cell memory against SARS-CoV-2. CD3+ T-lymphocytes were analyzed using an Aquios CL flow cytometer with a monoclonal antibody mixture including anti-CD3-PC5 (clone UCHT1). Samples were stained with CD3-FITC/CD4-PE Simultest, and blasts were identified as high FSC cells, divided into CD4 (CD3+CD4+) and CD8 (CD3+CD4-) populations using KaluzaC software. SARS-CoV-2 nucleocapsid and spike proteins were tested, with pokeweed mitogen as a positive control for T cell blast formation.

B7-H7 (HHLA2) inhibits T-cell activation and proliferation in the presence of TCR and CD28 signaling

This study examines the inhibitory effects of B7-H7 (HHLA2) on T-cell activation and proliferation in the presence of TCR and CD28 signaling. The anti-CD3 antibody clone UCHT1 was used alongside OKT3 to evaluate T-cell activation. B7-H7, combined with UCHT1, induced expression of CD25, CD69, and PD-1 on T cells, with less pronounced coinhibitory effects compared to OKT3. Plate coating included anti-CD3 UCHT1, anti-CD3 OKT3, IgG control, B7.1 Fc chimera, and B7-H7 Fc chimera, demonstrating differential impacts on T-cell activation.

Background

The UCHT1 antibody binds to CD3 epsilon of the T cell receptor-CD3 complex found only on mature T cells and medullary thymocytes. An interaction between T cells, UCHT1, and monocytes causes T-cell activation (mitogenesis) in vitro. On the other hand, the UCHT1 antibody blocks the unique association between the antigen receptor and CD3, and both the generation and function of cytotoxic T cells. After an initial dose of the UCHT1 antibody, T cells disappear from the circulation within minutes to hours. During treatment with the UCHT1 antibody, T cells bearing the usual array of surface molecules (CD2, CD4, and CD8) reappear in the peripheral blood circulation but these cells are devoid of CD3. After another 48 hours without the UCHT1 antibody, the normal array of surface molecules including CD3 appear again on all T cells. The selective removal of CD3 by internalization is thought to be the key mechanism of action of the UCHT1 antibody. Comodulation of the antigen receptor with CD3 explains the immunoblocking action of the UCHT1 antibody in vivo. The UCHT1 antibody is ineffective clinically if the CD3 target molecule is not modulated.

The UCHT1 antibody is effective for induction of immunosuppression and for treating initial and steroid-resistant rejections.
