Source: The anti-mouse CD3e monoclonal antibody (clone: 500A2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 500A2) specifically binds to the mouse T cell receptor CD3e.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD3e protein.

Heteroconjugated antibodies enhance lymphocyte-mediated tumour cell lysis in vitro and in vivo

T lymphocytes were activated in vivo by intraperitoneal injection of 50 ?g of SEB in C3H/HeJ mice and, after 18 h, spleen cells were prepared and cultured with 100 U/ml IL-2 for 3-7 days in the presence or absence of bispecific antibodies 500A2 × 96.5. The bispecific antibody 500A2 × 96.5 (anti-CD3 × anti-p97) was used to direct SEB-activated T lymphocytes against p97-positive melanoma cells. The melanoma target cell line CL62, which expresses the p97 antigen, was injected i.p. into C3H/HeJ mice to establish peritoneal tumors. The combination of in vivo SEB activation and in vitro IL-2 culture significantly enhanced the lysis of CL62 cells in the presence of bispecific antibodies. In vivo tumour neutralization experiments were performed by mixing CL62 tumour cells with SEB-activated T lymphocytes and bispecific antibodies before i.p. injection into C3H/HeJ mice.

The bispecific antibody 500A2 x 96.5 targets T-lymphocytes activated in vivo with staphylococcal enterotoxin B (SEB) against CL62 melanoma cells in vitro. We activated T-lymphocytes in vivo with SEB, expanded them in vitro with IL-2, and directed them against a tumour target with the BAb 500A2 x 96.5, specific for the murine CD3 antigen and the melanoma p97 antigen expressed by the CL62 tumour. C3H mice received SEB 50 micrograms intraperitoneally (i.p.). After 18 h mice were sacrificed and splenocytes extracted and either passed over a nylon wool column to isolate T-lymphocytes, or cultured in vitro for 3 to 7 days with 100 U ml-1 of IL-2. A 4-h chromium-release assay was used to assess the ability of T-lymphocytes to lyse the tumour target CL 62 in the presence or absence of the bispecific antibody 500A2 x 96.5. The addition of BAb significantly enhanced tumour lysis by SEB activated cells after a period of in vitro culture with IL-2.

Antitumor x anti-CD3 bifunctional antibodies redirect T-cells activated in vivo with staphylococcal enterotoxin B to neutralize pulmonary metastases

Antitumor x anti-CD3 (500A2) x anti-p97 (96.5) BFA. Antitumor x anti-CD3 bifunctional antibodies redirect T-cells activated in vivo with staphylococcal enterotoxin B to neutralize pulmonary metastases. Pulmonary metastases were counted 14 days following injections. SE-B alone induced a dose-dependent activation of T cells in vivo. The combination of SEB activation and BFA treatment significantly reduced pulmonary metastases.

Pulmonary metastases neutralization and tumor rejection by in vivo administration of beta glucan and bispecific antibody

To test for tumor neutralization, C3H/HeN mice were injected i.v. with Cl-62 melanoma cells and immediately treated with i.p. beta glucan and/or anti-CD3 (500A2) antibody. The bispecific antibody 500A2 x 96.5 was used to direct activated T lymphocytes against p97-positive melanoma cells. In vivo administration of beta glucan enhanced the therapeutic effect of bispecific antibody. Tumor rejection was observed in mice treated with the combination. The treatment significantly reduced pulmonary metastases.

Anti-CD3 x anti-tumor F(ab')2 bifunctional antibody activates and retargets tumor-infiltrating lymphocytes

The ability of intact and F(ab')2 anti-CD3 (500A2) x anti-p97 (96.5) BFA to induce activation of T lymphocytes in the presence of murine melanoma tumor cells (CL-62) expressing human melanoma-associated Ag (p97) was investigated in vitro and in vivo. These results demonstrate that F(ab')2 BFA can induce activation of cytotoxic lymphocytes only in the presence of relevant tumor cells, both in vitro and in vivo. That these activated lymphocytes can be redirected to lyse relevant tumor cells by the same BFA has important implications for the clinical application of BFA anti-tumor therapies. In vivo, the BFA was administered to mice with peritoneal tumors. The treatment led to tumor reduction in vivo.
