The anti-CD3 antibody clone 500A2 is recommended for mouse T cell identification in multicolor flow cytometry panels. For surface staining, antibodies were used at predetermined optimal dilutions in FACS buffer for 20 min at 4°C in the dark. Cells were washed twice with FACS buffer and fixed in 1% paraformaldehyde. Dead cells were excluded using a viability dye such as Zombie Aqua. Flow cytometry data were acquired on a BD FACSCanto II and analyzed using FlowJo software.

anti-Mouse CD3e 500A2 was used for flow cytometric identification of CD8+ T cells in tumor infiltrates. Cells were incubated with recommended dilutions of antibodies against cell surface proteins in PBS containing 0.1% fixable viability dye for 30 minutes at 4°C. For labelling of cells in whole blood, 50-100 μl of PB were stained with antibodies specific for surface molecules for 30 minutes at 4°C, followed by incubation for 10 minutes with 2 ml of 1X BD FACS lysing solution to remove erythrocytes. Cytotoxic function of CD8+ effector T-cells was assessed by flow cytometric analysis of degranulation capacity, as measured by CD107a expression on the cell surface, GzmA and GzmB expression or IFNγ production. Flow cytometry data were analyzed using FlowJo software version 10.

The clone 500A2 anti-CD3 antibody is suggested for gating T cells in mouse flow cytometry experiments. Antibody staining was performed in PBS with 2% FBS and 0.09% sodium azide for 30 min on ice. Samples were acquired on a BD LSRFortessa and data analyzed with FlowJo v10. Compensation was performed using single-stained controls. Viability was assessed with propidium iodide.

Anti-CD3 (clone 500A2) antibody was utilized in the flow cytometry panel for T lymphocyte subset analysis. Cells were stained with fluorochrome-conjugated antibodies for 20 minutes at room temperature. After washing, samples were run on a FACSCalibur flow cytometer. Data analysis included compensation matrices derived from single-color controls. The protocol ensures minimal spectral overlap in multicolor setups.

Deep metabolic profiling of immune cells by spectral flow cytometry reveals dynamic changes in mitochondrial function during activation

500A2 (AF700-conjugated) was employed for T cell phenotyping in spectral flow cytometry to track activation states. Isolated splenocytes were stained with a panel of antibodies including anti-CD3 (500A2) for 30 min at 4°C. Mitochondrial membrane potential was assessed using TMRE dye concurrently. Data were collected on a Cytek Aurora spectral cytometer and unmixing performed with SpectroFlo software. Activation-induced metabolic shifts were quantified in CD3+ populations over time.
