Source: The anti-human CD3 monoclonal antibody (clone: UCHT1) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: UCHT1) specifically binds to the human T cell receptor CD3.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human CD3 protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.

In vivo DNA-launched bispecific T cell engager targeting IL-13R?2 limits tumor growth in a humanized mouse model

This study evaluates a DNA-launched bispecific T cell engager (BiTE) targeting IL-13R?2 in a humanized NSG mouse model engrafted with human PBMCs. Flow cytometry analysis utilized anti-human CD45 (HI30), anti-human CD3 (UCHT1), anti-human CD4 (OKT4), anti-human CD8 (SK1), and anti-human PD-1 (EH12.2H7) antibodies. Tumor volumes were measured twice weekly using calipers, calculated as (length × width?)/2. The IL-13R?2 BiTE, delivered via hydrodynamic injection of plasmid DNA, significantly reduced tumor growth compared to controls.

SCENITH: A Flow Cytometry-Based Method to Functionally Profile Energy Metabolism with Single-Cell Resolution

This study introduces SCENITH, a flow cytometry-based method to profile energy metabolism at single-cell resolution. Immune cell phenotyping was performed using antibodies including anti-CD45 (HI30), anti-CD3 (UCHT1), anti-CD4 (RPA-T4), anti-CD8 (SK1), anti-CD11b (ICRF44), and anti-CD19 (HIB19). Mice were injected with 2-deoxyglucose to inhibit glycolysis, and spleens were processed into single-cell suspensions for analysis. SCENITH measures protein synthesis rates as a proxy for cellular energy metabolism, validated in both mouse and human immune cells.

An IgG-based bispecific antibody for improved dual targeting in vivo

This study investigates an IgG-based bispecific antibody in NSG mice engrafted with human PBMCs and tumor cells. T cells were stained with anti-CD3 (UCHT1) and anti-CD8 (SK1) antibodies for flow cytometry analysis. Tumor growth was monitored via bioluminescence imaging using an IVIS Spectrum system. The bispecific antibody was administered intravenously at 0.5 mg/kg weekly, with peripheral blood analyzed for T cell activation and cytokine release post-treatment.

In vitro Characterization of the Anti-PD-1 Antibody Nivolumab, BMS-936558, and in vivo Toxicology in Non-Human Primates

This study characterizes the anti-PD-1 antibody nivolumab in vitro and evaluates its toxicology in cynomolgus monkeys. Flow cytometry was performed using anti-CD3 (UCHT1), anti-CD4 (L200), and anti-CD8 (RPA-T8) antibodies. Monkeys were dosed with nivolumab at 10, 50, or 100 mg/kg weekly for 4 weeks, with peripheral blood samples collected at multiple time points. Nivolumab binding to PD-1 was confirmed on activated T cells, and no significant toxicity was observed at any dose level.

Preclinical evaluation of light-activatable, bispecific anti-human CD3 antibody conjugates as anti-ovarian cancer therapeutics

This study evaluates light-activatable UCHT1-folate conjugates in nude mice bearing ovarian cancer xenografts. Flow cytometry confirmed T cell binding using anti-CD3 (UCHT1) antibody. Tumors were irradiated with 690 nm light to activate the conjugate, and tumor volumes were measured every 3 days using calipers. The conjugate significantly reduced tumor growth compared to non-irradiated controls.
