Macrophage Migration Inhibitory Factor protects cancer cells from immunogenic cell death and impairs anti-tumor immune responses
Balogh, K. N., et al. PLoS One. 2018 Jun 4;13(6):e0197702. doi: 10.1371/journal.pone.0197702. PMID: 29864117
Beginning two days before 4T1 cell tumor implantation, mice were treated with intraperitoneal injection of an initial dose of 200ug/mouse of anti-CD4 (clone GK1.5, BioXCell) and anti-CD8 (clone 2.43, BioXCell) antibodies in PBS, followed by similar dosing with 100ug/mouse every 4 days throughout the course of tumor growth. ...The MIF KD tumors also contained significantly fewer CD4+ T cells by percent. ...However, no significant difference was observed in the total number of CD4+ T cells. ...Interestingly, both populations of tumor-infiltrating T cells in the MIF KD tumors were more activated, as exhibited by the ability of both CD8+ and CD4+ T cells to express IFNgamma after ex vivo re-stimulation. ...Therefore, we depleted the CD4 and CD8 T cells simultaneously in the same mice.

TGF? Is a Master Regulator of Radiation Therapy-Induced Antitumor Immunity
Vanpouille-Box, C., et al. Cancer Res. 2015 Jun 1;75(11):2232-42. doi: 10.1158/0008-5472.CAN-14-3511. PMID: 25858148
Depletion of CD4+ and CD8+ T cells was achieved by injecting GK1.5 or 2.43 mAb (BioXCell) given i.p. at 100 ?g/mouse on 3 consecutive days, starting at day 10, and was maintained by weekly injections. ...Briefly, TDLN cells were stained with anti-mouse CD4-PE and anti-mouse CD8-FITC (eBioscience), fixed, permeabilized (Foxp3 Fixation/Permeabilization Concentrate and Diluent kit, eBioscience), and stained with goat anti-phospho-Smad2/3 (Ser 423/425) followed by APC-labeled donkey anti-goat IgG (Santa Cruz Biotechnology). ...Images were obtained using a Leica SNC400F fluorescence slide scanner. CD4 and CD8 T cells were counted in five randomly selected (×200) fields in each tumor. ...TIDC, defined as CD45+CD11c+MHC-II+ cells, was analyzed at day 22 for expression of activation markers CD40 and CD70, which plays a critical role in controlling priming versus tolerance induction of CD8+ T cells. ...Consistent with this, depletion of CD4+ or CD8+ T cells abrogated regression of the irradiated 4T1 tumors and restored the number of lung metastases in mice treated with TGF? inhibition + RT.

Memory programming in CD8(+) T-cell differentiation is intrinsic and is not determined by CD4 help
Kim, J., et al. Nat Commun. 2015 Aug 14:6:7994. doi: 10.1038/ncomms8994. PMID: 26272364
Stimulation of CD8+ T cells in the absence of CD4+ T-cell help is an important constraint on the quantity and quality of the CD8+ T-cell response, resulting in defects in memory expansion of activated CD8+ T cells. ...The general consensus is that CD4 help delivered during CD8+ T-cell priming encodes a programme in the activated CD8+ T cells to generate memory cells. ...Nevertheless, what is the fundamental role of CD4+ T cells in preventing memory impairment of CD8+ T cells remains to be elucidated. ...However, the reliance on contrived approaches to create helper deficiency, such as CD4 depletion and the use of major histocompatibility complex (MHC) II- or CD4-deficient mice, and the paucity of antigen-specific CD8+ T cells expanded after helper-deficient activation limit extrapolating these results to physiological situations. ...However, this immunodominance is CD4+ T-helper cell-dependent.

IL-27 Signaling Is Crucial for Survival of Mice Infected with African Trypanosomes via Preventing Lethal Effects of CD4+ T Cells and IFN-?
Liu, G., et al. PLoS Pathog. 2015 Jul 29;11(7):e1005065. doi: 10.1371/journal.ppat.1005065. PMID: 26222157
Either depletion of CD4+ T cells, resulting in a dramatically reduced secretion of IFN-?, or neutralization of IFN-?, prevented the early mortality of infected IL-27R-/- mice with a significantly reduced inflammatory response and a major amelioration of the liver pathology. ...Thus, our data identify IL-27 signaling as a novel pathway to prevent the early mortality via inhibiting hyperactivation of CD4+ Th1 cells and their excessive secretions of IFN-? during experimental infection with extracellular protozoan parasites African trypanosomes. ...More recent studies have suggested that IL-27 also has the function to inhibit immunopathology via downregulation of active CD4+ T cells during infections, particularly with intracellular protozoan parasites. ...However, the precise mechanism of CD4+ T cell-mediated immunopathogenesis in the absence of IL-27 signaling still remains incompletely understood. ...A limited and similar percentage and absolute number of CD4+ T cells from uninfected wild-type and IL-27R-/- mice produced IFN-? after 12 h stimulation with Cell Stimulation Cocktail (containing PMA, ionomycin, and protein transport inhibitors).

Other antibody clones, including anti?PD-1/MAb RMP1-14, anti-CD8/MAb 2.43, and anti-CD4/MAb GK1.5, were purchased from BioXCell and were verified to have <2 EU/mg endotoxin levels, ?95% purity, and ?5% high-molecular-weight species before in vivo use. ...Anti-SEMA4D and anti?CTLA-4 demonstrated independent and combinatorial effects on Teff to Treg ratios as determined by FACS analysis (Teff = CD8+/CD69+; Treg = CD4+/CD25+; Fig. 5C and Supplementary Fig. S5A). ...Tumor-associated M2 macrophages can interact with lymphocytes to promote tumor growth; for example, TAMs can regulate the influx of CD4+ Tregs and can inhibit cytotoxic CD8+ T-cell responses through several mechanisms. ...Spleens were dissociated into small pieces and the tissue homogenate was pressed through a 0.07-mm cell strainer, followed by RBC lysis (Bioscience) for 5 minutes at room temperature. ...Splenocytes and TILs were assayed by Cytometric Bead Array (CBA) cytokine analysis.
