Source: The anti-human CD4 monoclonal antibody (clone: OKT4A) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: OKT4A) specifically binds to human and non-human primate CD4.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human and non-human primate CD4 protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.
Endotoxin: < 1 EU per 1 mg of the protein by the LAL method.

Background
The recombinant OKT4A antibody binds to the human CD4 protein (cluster of differentiation 4), a glycoprotein and the co-receptor for the T-cell receptor (TCR). The CD4 protein is expressed on the surface of immune cells such as helper T cells, monocytes, macrophages, and dendritic cells.

A rhesus recombinant OKT4A antibody (anti-human CD4) was administered at 50 mg/kg intravenously to deplete CD4+ T cells in SIV-infected macaques. Flow cytometry showed maximal depletion within one week, with significant loss of CD28+CD95− CD4+ T cells and SIV-specific responses, persisting for at least 8 weeks, without affecting viral load in elite controllers.

The OKT4A antibody (anti-human CD4) was studied for its immunosuppressive effects in nonhuman primate models of graft rejection and autoimmune disease. Humanized gOKT4A-4 showed reduced immunogenicity compared to murine OKT4A, with computer modeling guiding humanization to retain binding activity, and preliminary clinical trials indicated no cytokine release syndrome or CD4+ T cell depletion.

Infection of Brain Microglial Cells by Human Immunodeficiency Virus Type 1 Is CD4 Dependent

The OKT4A antibody (anti-human CD4) was used to block HIV-1 infection in cultured microglial cells. Pretreatment with OKT4A significantly reduced infection by HIV-1 strains AD87(M) and BaL, as measured by reverse transcriptase and p24 antigen levels, confirming CD4-dependent viral entry in brain-derived microglial cells via flow cytometry.

The OKT4 antibody (anti-human CD4), but not OKT4A (anti-human CD4), co-immunoprecipitated a specific gp160-CD4 complex in the endoplasmic reticulum of coexpressing cells. OKT4A’s inability to bind this complex confirmed its distinct epitope specificity, demonstrating that gp160-CD4 interaction prevents CD4 maturation and surface expression, as assessed by flow cytometry.

The OKT4A antibody (anti-human CD4) was administered to three longtailed macaques with experimental allergic encephalomyelitis (EAE). Treatment reversed CD4+ and CD8+ cell depletion, reduced brain lesion inflammation, and prolonged survival, suggesting OKT4A’s potential in treating human demyelinating diseases by modulating CD4+ T cell activity.
