Source: The anti-human CD4 monoclonal antibody (clone: 13B8.2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 13B8.2) specifically binds to human CD4.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human CD4 protein.

Background

The recombinant 13B8.2 antibody binds to the human CD4 protein (cluster of differentiation 4), a glycoprotein and the co-receptor for the T-cell receptor (TCR). The CD4 protein is expressed on the surface of immune cells such as helper T cells, monocytes, macrophages, and dendritic cells.

Peripheral blood mononuclear cells (PBMCs) from 12 donors were stained with anti-CD4–PE (clone 13B8.2) antibody, along with anti-CD27–PE-Cy5 (clone O323), anti-CD25–eFluor450 (clone BC96), anti-CD31–PE-Cy7 (clone WM59), and anti-CD45RA–FITC (clone JS-83) to identify recent thymic emigrants (RTEs), mature naive T cells, and naive Tregs in the CD4+ lineage. The study explored the plasticity of Th2, Th1, Th17, and Treg cells, noting that Th2 cells can adopt a Th1-like phenotype, while Th1 and Th17 cells can transition to Th2 in helminth-infected mice, and Th17 cells can acquire regulatory features like IL-10 production under IL-12 or IL-27 influence.

Macrophage infection via selective capture of HIV-1-infected CD4+ T cells

The study used the 13B8.2 antibody (anti-human CD4, 10 μg/ml) as an inhibitor of HIV-1 Env-receptor interactions, alongside soluble CD4, Tak-779, 2G12, and T20. The results suggest that macrophage uptake of HIV-1-infected CD4+ T cells contributes to the macrophage reservoir and viral dissemination in vivo, particularly after transmission or during massive CD4+ T cell death in lymphoid tissues. Detection of viral DNA or proteins in phagocytes may indicate uptake of infected immune cells rather than direct infection.

In vitro antitumoral activity of baculovirus-expressed chimeric recombinant anti-CD4 antibody 13B8.2 on T-cell lymphomas

A baculovirus-expressed chimeric recombinant IgG1 antibody (rIgG1) with human Cγ1 and Cκ domains, derived from the murine anti-CD4 monoclonal antibody 13B8.2, was tested for antitumoral activity against CD4+ T-cell lymphomas. The 13B8.2 antibody, specific for the CDR3-like loop of CD4, inhibited HIV-1 replication and T cell proliferation in vitro and showed promising results in CD4+ lymphoma cell lines, leading to plans for in vivo preclinical studies in a murine xenograft model.

The 13B8.2 monoclonal antibody, targeting the CD4 region critical for receptor dimerization, prevented apoptosis in HIV-infected cells without inhibiting viral replication. The study highlighted high levels of HIV replication and immune system activation in vivo, contributing to enhanced apoptosis in both infected and bystander CD4 cells. Fas signaling was noted as a potential contributor to T-cell depletion in HIV-1-infected patients.

Differential CD4-dependent inhibition of JNK but not Erk-2 activities by anti-CD4 monoclonal antibodies

The study observed differential inhibition of JNK but not Erk-2 activities by the anti-CD4 monoclonal antibody 13B8.2 in Jurkat T cells. In vivo experiments in mouse models showed that administration of 13B8.2 led to selective inhibition of JNK activity in T cells, demonstrating that CD4 ligation modulates T cell activation through differential kinase inhibition.
