Background:

The recombinant OKT4 antibody binds to the human CD4 protein (cluster of differentiation 4), a glycoprotein and the co-receptor for the T-cell receptor (TCR). The CD4 protein is expressed on the surface of immune cells such as helper T cells, monocytes, macrophages, and dendritic cells.

The OKT4 antibody (anti-human CD4) was used to identify a deficiency in the CD4 epitope in patients with normal CD4+ T cell counts, as detected by other anti-CD4 monoclonal antibodies. This study linked a single amino acid substitution in the V3 domain of CD4 to the OKT4 epitope deficiency, relevant to understanding HIV pathogenesis as CD4 is the HIV receptor.

The OKT4 antibody (anti-human CD4) was used in flow cytometry to analyze surface antigen changes in activated T lymphocytes. The study observed a shift in the OKT4:OKT8 ratio from 60:40 to 40:60 in cultured T lymphoblasts, suggesting a possible phenotype change from OKT4+ to OKT8+ cells, while OKT4 expression remained stable.
