Background:

The recombinant 13B8.2 antibody binds to the human CD4 protein (cluster of differentiation 4), a glycoprotein and the co-receptor for the T-cell receptor (TCR). The CD4 protein is expressed on the surface of immune cells such as helper T cells, monocytes, macrophages, and dendritic cells.

The chimeric recombinant anti-CD4 antibody 13B8.2, expressed in baculovirus, was tested for antitumoral activity against T-cell lymphomas using flow cytometry. The 13B8.2 antibody induced apoptosis in CD4+ Jurkat cells, as confirmed by Annexin V staining, and significantly reduced CD4+ T cells in treated lymphoma cell lines. Cell viability was assessed after 48 hours using MTT assays, and the efficacy of 13B8.2 was compared with other anti-CD4 antibodies in vitro.

The anti-CD4 monoclonal antibody 13B8.2, targeting the CDR3-like region of CD4, was used to assess HIV-1 envelope glycoprotein-induced apoptosis via flow cytometry. Preincubation with 13B8.2 significantly reduced apoptosis in HIV-1-infected T cells and blocked CD4 crosslinking by gp120. The study also demonstrated that 13B8.2 inhibits syncytium formation in vitro, with cell surface CD4 expression quantified using fluorescently labeled antibodies.

The 13B8.2 antibody (anti-human CD4 antibody) was used to detect CD4 expression on transfected HeLa cells by flow cytometry to study HIV-1 envelope-mediated cell fusion. Mutations in the CD4 cytoplasmic tail abolished the inhibitory effect on cell fusion, while flow cytometry confirmed that CD4 surface expression remained unchanged. HIV-1 envelope glycoprotein binding was assessed through co-culture experiments.

CD4 is required for inhibition of human immunodeficiency virus type 1 replication by antibodies that bind to the immunoglobulin CDR3-like region in domain 1 of CD4

The 13B8.2 antibody effectively blocked HIV-1 provirus transcription without significantly altering CD4 surface levels. Inhibition of HIV-1 entry was confirmed using syncytium formation assays.
