Background
The rat anti-mouse CD8a monoclonal antibody YTS 169.4 (rat IgG2b kappa) reacts with the mouse CD8a protein (T-cell surface glycoprotein CD8 alpha chain) encoded by the mouse CD8A gene that encodes the CD8a chain of the dimeric CD8 protein. The mouse CD8 protein is primarily responsible for cell-mediated immune defense and T-cell development. CD8A has been widely reported as a potential prognosis and diagnostic marker for several diseases, such as inflammatory disorders and tumors. It was shown that the anti-mouse CD8a rat monoclonal antibody (clone number: YTS 169.4) can be used for depleting activity in vivo.
Our recombinant YTS 169.4 antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse CD8a monoclonal antibody (hybridoma clone name or number: YTS 169.4).
1、Differential effectiveness of anti-CD8 treatment on ongoing graft-versus-host reactions in mice.
Noort, W. A., et al. Transpl Immunol. 1996 Sep;4(3):198-202. doi: 10.1016/s0966-3274(96)80017-7. PMID: 8893449
Mice were treated significantly with a single optimally depleting dose of rat anti-CD8 (activity of anti-mouse CD8a antibody) (YTS 169.4) or untreated. ...Symptoms of GVHD became obvious 6 days after reconstitution, and mortality started at day 8. ...The mutual influence of CD4+ and CD8+ T cells in the development of GVHD becomes apparent from our data, and demonstrates that GVHD lethality can be caused by CD8+ T cells as well as by CD4+ T cells. ...Analysis of T cell subsets in the spleen during graft-versus-host (GVH) reactions in a fully allogeneic mouse strain combination demonstrated that first CD4+ T cells become activated, and initiate the GVH reaction. ...Here we show that anti-CD8 treatment (anti-mouse CD8a antibody for animal model) on day +3 resulted in a significant increase in survival, while early treatment (day -1 or day +1) did not.

2、Depletion of murine CD8+ T cells in vivo decreases pulmonary clearance of a moderately virulent strain of Cryptococcus neoformans.
Mody, C. H., et al. J Lab Clin Med. 1993 Jun;121(6):765-73. PMID: 7685044
Mice depleted of CD8+ T cells by monoclonal antibody (anti-mouse CD8a for cancer research) (YTS 169.4) injections were infected with the moderately virulent cryptococcal strain 613D by the intratracheal route, which mimics natural pulmonary infection. ...To ensure adequacy of depletion, T cell subsets were enumerated by flow cytometry in blood, spleen, lymph node, and lung. ...Specific elements of host defense were measured by clearance of cryptococci in vivo and by the response to cryptococcal antigens, both in vivo by delayed-type hypersensitivity (DTH) and in vitro by lymphocyte proliferation. ...We found that depletion of CD8+ T cells decreased pulmonary clearance of C. neoformans. CD8 depletion abrogated DTH without affecting antigen recognition or lymphocyte proliferation in vitro. ...Host defense mechanisms to the important fungal pathogen Cryptococcus neoformans are complex and incompletely understood.
