Background:
The recombinant OKT8 antibody binds to the human CD8 protein (cluster of differentiation 8), a type I transmembrane glycoprotein and the co-receptor for the T-cell receptor (TCR). Along with the TCR, the CD8 co-receptor plays a role in T cell signaling and aiding with cytotoxic T cell-antigen interactions. The CD8 protein is predominantly expressed on the surface of cytotoxic T cells, but can also be found on natural killer cells, cortical thymocytes, and dendritic cells. The CD8 molecule is a marker for cytotoxic T cell population.

The OKT8 antibody (anti-human CD8a) was used in flow cytometry to confirm specific transduction of CD8+ T cells in a mouse model. OKT8 staining showed high transduction efficiency in the CD8+ population compared to CD4+ cells, with OKT8-positive cells gated for sorting, enhancing tumor killing in transduced cells.

The OKT8 antibody (anti-human CD8a) was used in flow cytometry to identify cytotoxic T cells lacking the OKT8 antigen. OKT8-negative CTL clones exhibited normal cytolytic activity, indicating that the absence of OKT8 did not impair function, with OKT8 serving as a marker for conventional CTL.

The OKT8 antibody (anti-human CD8a) identified a subset of NK and K cells expressing the OKT8 antigen, detected in 20-30% of NK cells via flow cytometry. OKT8-positive NK cells showed cytotoxic activity similar to T cells, and OKT8 was used to isolate this subset for further analysis.

The OKT8 antibody (anti-human CD8a) was analyzed for binding to CD8+ T cells via flow cytometry at various temperatures. OKT8 showed endothermic binding characteristics, with increased fluorescence intensity at higher temperatures, and its affinity was calculated from flow cytometry data for cytometric detection.
