The rat anti-mouse CD8a monoclonal antibody YTS 169.4 (rat IgG2b kappa) reacts with the mouse CD8a protein (T-cell surface glycoprotein CD8 alpha chain) encoded by the mouse CD8A gene that encodes the CD8a chain of the dimeric CD8 protein. The mouse CD8 protein is primarily responsible for cell-mediated immune defense and T-cell development. CD8A has been widely reported as a potential prognosis and diagnostic marker for several diseases, such as inflammatory disorders and tumors. It was shown that the anti-mouse CD8a rat monoclonal antibody (clone number: YTS 169.4) can be used for depleting activity in vivo.
Our recombinant YTS 169.4 antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse CD8a monoclonal antibody (hybridoma clone name or number: YTS 169.4).

Influenza-Activated ILC1s Contribute to Antiviral Immunity Partially Influenced by Differential GITR Expression
Vashist, N., et al. Front Immunol. 2018 Mar 22:9:505. doi: 10.3389/fimmu.2018.00505. PMID: 29623077
Monoclonal anti-mouse CD8 (YTS 169.4, BioXCell, USA) and CD4 antibodies (GK1.5, BioXCell, USA) were used to deplete CD8 and CD4 T cells in vivo, respectively, at 0 and 3 days post infection (dpi) (200 ?g/animal in 200 ?l PBS, i.p.). ...The activation status of infected BMDCs was subsequently assessed by their expression levels of specific maturation markers (CD40, CD80, CD86, and MHC cl. II). ...As expected, H1N1 infection induced enhanced expression of CD40, CD80, CD86, and MHC cl. II on BMDCs (Figure 4E). ...CD8 T cells are crucial in clearing influenza infection in the absence of antibody responses. ...Thus, the H1N1 PR8 in vitro infection model was exploited to assess whether ILC1s cross-talk with CD8 T cells (Gating strategy; Figure S9A in Supplementary Material).

Myeloid Cell Arg1 Inhibits Control of Arthritogenic Alphavirus Infection by Suppressing Antiviral T Cells

The following antibodies from Bio X Cell were used for depletion studies: rat IgG2b control antibody (anti-KLH, clone LTF-2), CD4-depleting antibody (rat IgG2b; clone YTS 191), and CD8-depleting antibody (rat IgG2b; clone YTS 169.4). ...To investigate this hypothesis, WT and LysMcre;Arg1F/F mice were treated with anti-CD4 and anti-CD8? antibodies, or an isotype control antibody, on days 7 and 12 pi to deplete CD4+ and CD8+ T cells. ...Administration of anti-CD4 and anti-CD8? antibodies depleted ? 98% of both CD4+ and CD8+ T cell subsets as demonstrated by staining spleens for CD3, CD4, and CD8?. ...After gating on lymphocytes, CD8+ T cells were identified by staining positive for CD3 and CD8; CD4+ T cells were identified by gating on CD3+CD8- cells followed by gating on CD3+CD4+ cells. ...A subset of CD4+ T cells produced both IFN-? and IL-10, whereas CD8+ T cells produced IFN-? but very little IL-10.

For depletion of CD8+ T cells, mice were injected at day 15 and 17 post?bone marrow transplantation with 250 ?g of anti-CD8 antibody (YTS 169.4 clone, BioXcell) followed by 100 ?g every week. ...Flow cytometric analysis of CD4+ and CD8+ T cell frequencies in the spleen of chimeric mice at day 55 following anti-CD8? (YTS 169.4) or isotype control treatment. ...The lymphoproliferative disease in these mice is caused primarily by expansion of CD8+ T cells that even progress to leukaemia. ...Because CD4-Cre-mediated TR2-ablation resulted in a strong thymic phenotype including reduction of single positive CD8+ T cells, abrogation of NK T cell development, and enhanced negative selection [11],[12], we investigated the extent and consequences of induced TR2 ablation in the thymus. ...Flow cytometric analysis of TR2 expression by CD4+ and CD8+ T cells from peripheral blood after 5 mo postthymectomy and tamoxifen treatment.
