Background
The rat anti-mouse CD8a monoclonal antibody YTS 169.4 (rat IgG2b kappa) reacts with the mouse CD8a protein (T-cell surface glycoprotein CD8 alpha chain) encoded by the mouse CD8A gene that encodes the CD8a chain of the dimeric CD8 protein. The mouse CD8 protein is primarily responsible for cell-mediated immune defense and T-cell development. CD8A has been widely reported as a potential prognosis and diagnostic marker for several diseases, such as inflammatory disorders and tumors. It was shown that the anti-mouse CD8a rat monoclonal antibody (clone number: YTS 169.4) can be used for depleting activity in vivo.
Our recombinant YTS 169.4 antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse CD8a monoclonal antibody (hybridoma clone name or number: YTS 169.4).
The in vivo grade recombinant anti-mouse CD8a mouse IgG2a monoclonal antibody was produced in mammalian cells.

For in vivo depletion, anti-CD8 (YTS 169.4, 100 ?g) was administered intraperitoneally on days -3 and 0 relative to immunization. Flow cytometry was used to quantify retinal infiltration and confirm CD8+ T cell depletion. Chronic experimental autoimmune uveoretinitis (EAU) was induced in B10.RIII mice by immunization with IRBP peptide. Effector memory CD8 T cells were found to expand in the chronic phase of EAU. Depletion of CD8+ T cells reduced retinal inflammation but did not eliminate disease progression.

Site-restricted persistent cytomegalovirus infection after selective long-term depletion of CD4+ T lymphocytes.

The mAbs used for depletion were anti-L3T4 (clone YTS 191.1) and anti-Lyt-2 (clone YTS 169.4) at a dose of 1 mg per mouse, administered intraperitoneally. Selective depletion of CD4+ T lymphocytes was performed to study persistent cytomegalovirus infection. Viral titers were measured in salivary glands and lungs of infected mice. CD4+ T cell depletion led to site-restricted viral persistence in the salivary glands. The study demonstrated the critical role of CD4+ T cells in controlling cytomegalovirus infection.

In vivo depletion of murine CD8 positive T cells impairs survival during infection with a highly virulent strain of Cryptococcus neoformans.

Mice were injected intraperitoneally with 100 ?g of anti-CD8 monoclonal antibody (YTS 169.4) to deplete CD8+ T cells in vivo. Survival was monitored in mice infected with a highly virulent strain of Cryptococcus neoformans. Lung fungal burden was assessed by quantitative culture. Flow cytometry confirmed the depletion of CD8+ T cells in peripheral blood. CD8+ T cell depletion significantly reduced survival compared to control mice.

The rat anti-mouse Lyt-2 monoclonal antibody (YTS 169.4) was used to eliminate T-cell subsets in vivo in ATX mice. Monoclonal antibodies were administered intraperitoneally to deplete specific T-cell populations. The efficacy of T-cell depletion was confirmed by flow cytometry of spleen cells. Skin graft survival was prolonged in mice treated with anti-T-cell antibodies. The study demonstrated the therapeutic potential of monoclonal antibodies in immunosuppression.

CD4 monoclonal antibody pairs for immunosuppression and tolerance induction.

Anti-CD8 (YTS 169.4) and anti-CD4 (YTS 191.1) antibodies were administered intraperitoneally to achieve immunosuppression in vivo. The combination of monoclonal antibodies was tested for its ability to induce tolerance to skin grafts. Flow cytometry was used to monitor T-cell depletion in peripheral blood and lymphoid organs. Long-term graft survival was achieved in mice treated with both antibodies. The study highlighted the synergistic effect of anti-CD4 and anti-CD8 antibodies in tolerance induction.
