For in vitro stimulation, naive CD4+ T cells were activated with plate-bound anti-CD3 (5 μg/mL; clone 145-2C11) and soluble anti-CD28 antibody(2 μg/mL; clone 37.51) for 3 days. Cells were stained with antibodies against CD4, CD25, and CD69 to assess activation status by flow cytometry. The expression of HDAC3 was knocked down using siRNA in CD4+ T cells. Cholesterol efflux was measured using a fluorometric assay. Flow cytometry data were analyzed using FlowJo software.

CBM complex-deficient Treg cells were adoptively transferred into tumor-bearing mice. Tumor growth was measured twice weekly. Immune checkpoint therapy was administered intraperitoneally. Tumor-infiltrating lymphocytes were analyzed by flow cytometry.
