We next use the antibody fragment F(ab')2 targeting PV1 (anti-mouse CD28 antibody) to assess the dual effects of rapid kidney filtration and PV1 targeting on kidney-selective targeting. Ex vivo fluorescence imaging results demonstrated that after rapidly accumulating in kidneys at 4 h, PV1-targeted F(ab')2 was continually retained in the kidney at 24 h, whereas the isotype control F(ab')2 underwent urinary elimination with significantly reduced signaling in the kidney. Our findings suggest that PV1-targeted F(ab')2 might be useful as a drug carrier for renal targeting and highlight the importance of affinity optimization for tissue targeting antibodies. We investigated how targeting plasmalemma vesicle-associated protein (PV1), a protein found in the endothelial caveolae of lungs and kidneys, can promote accumulation in these organs. Using ex vivo fluorescence imaging, we show that intravenously administered αPV1 antibodies localize to mouse lungs and kidneys. Ex vivo imaging of lung and kidney shows higher fluorescence intensity in the lungs and kidneys of mice injected with 2 mg/kg of Alexa Fluor 680-labeled αPV1 monoclonal (a) and bispecific (b) antibodies compared with respective Alexa Fluor 680-labeled isotype controls at 24 h after intravenous injection. Quantification of fluorescence intensity from ex vivo imaging was normalized to the skeletal muscle (mean + SD) for αPV1 vs. isotype mAb (c) and for the bispecific αPV1 construct and bispecific isotype (d). Brackets indicate fold change in the fluorescence intensity of αPV1 to isotype control in lungs and kidneys.

In addition, administration of anti-PV-1 antibody during capillary flow assays resulted in a significant inhibition of lymphocyte transmigration through the endothelial cell layer, whereas rolling and adhesion were unaffected. In vivo blockage of PV-1 by an antibody in acute peritonitis and air pouch model resulted in a significant decrease in the number of migrating leukocytes. For double stainings, cells were first exposed to anti–PV-1 monoclonal antibody (mAb) 174/2 (mIgG1), followed by Alexa 488 conjugated goat anti–mouse IgG1 secondary antibody. For the last 20 minutes, anti-PV-1 mAb 174/2 or negative control mAb 3G6 was added at a concentration of 10 μg/mL. Plasmalemmal vesicle associated protein-1 (PV-1) is a marker of blood-brain barrier disruption in rodent models.

Our results confirm that PV-1 is preferentially induced in the endothelium of mouse brain tumors and acute ischemic brain tissue and corresponds to blood-brain barrier disruption in a fashion analogous to human patients. Characterization of PV-1 expression in mouse brain is the first step towards developing PV-1 as a surrogate marker of BBB disruption in vivo. We demonstrate that PV-1 is selectively upregulated in mouse blood vessels recruited by brain tumor xenografts at the RNA and protein levels, but is not detected in non-neoplastic brain. Additionally, PV-1 is induced in a mouse model of acute ischemia. Mouse embryonic stem cell-derived blood-brain barrier model: applicability to studying antibody triggered receptor mediated transcytosis.

Immunoblotting using the anti-hPV1C antibody (PV1, two top panels) on PMA-treated or control ECs. The two top panels represent different exposure times of the same PV1 blot. The earliest time point PV1 protein was detected at was between 6 and 12 h (bottom panel). Immunoblotting with anti–β-actin mAb on the same membrane shows loading (Actin). Caveolin-1 levels remained the same during PMA treatment (Cav1). No PV1 was detected in these cells whereas β-actin (Actin, middle panel) and caveolin 1 (Cav1, bottom panel) are readily detectable on the same membrane. PMA-induced PV1 upregulation is PMA specific and does not require the kinase activity of PKC in ECs.
