The CD28.2 antibody (BD Biosciences), conjugated to PE, was used to stain peripheral blood T lymphocytes from healthy donors, isolated via Ficoll density gradient centrifugation. Dual-color flow cytometry with CD3-FITC on a FACSCalibur showed significantly higher CD28 expression on CD4+ T cells compared to CD8+ T cells, with data analyzed using CellQuest software.

The CD28.2 antibody, conjugated to APC, assessed CD28 expression on chronic lymphocytic leukemia (CLL) B cells from peripheral blood, stained with CD19 and CD5. Mean fluorescence intensity (MFI) quantification on gated B cells, using isotype controls to minimize non-specific staining, showed reduced CD28 expression correlating with disease progression.

Flow cytometric detection of CD28 and CTLA-4 in rheumatoid arthritis synovial fluid cells
Liu, M. F., et al. J Rheumatol. 2001 Dec;28(12):2600-2607. PMID: 11754362
Synovial fluid cells from rheumatoid arthritis (RA) patients, isolated within 4 hours post-arthrocentesis, were stained with CD28.2-FITC and CTLA-4-PE for dual-staining flow cytometry on a FACScan, collecting at least 10,000 events. CD28 expression was downregulated on synovial T cells compared to peripheral blood.

Flow cytometric assay for CD28-mediated T cell activation using CD28.2
Riley, J. L., et al. Clin Cancer Res. 1999 Jul;5(7):1853-1861. PMID: 10417773
The CD28.2 antibody, conjugated to PE-Cy5, was used to stimulate Jurkat T cells with anti-CD3 for 48 hours, followed by flow cytometric analysis on a FACStar Plus. Activation markers CD69 and CD25 were assessed, showing enhanced IL-2 production in CD28-stimulated T cells.

Flow cytometric characterization of CD28.2 on tumor-infiltrating T cells

The CD28.2 antibody stained tumor-infiltrating lymphocytes (TILs) from melanoma biopsies, isolated via enzymatic digestion, in a 6-color flow cytometry panel with CD3 and CD8. CD28 expression was reduced on tumor-specific CD8+ T cells, with data analyzed using FCS Express software for population gating.
