The anti-human PLVAP/PV-1 antibody used was clone MECA-32 (Santa Cruz Biotechnology, sc-19603). Human endothelial cells were isolated from fresh tissue samples. Flow cytometry was performed using a BD FACSCanto II. Cells were stained for CD31, CD144, and PLVAP to confirm endothelial identity. Data analysis was conducted with FlowJo software.

For mouse lung endothelial cells (MLEC), single-cell suspensions were stained with rat anti-mouse PLVAP/PV-1 antibody (clone MECA-32, BioLegend, #120506) and analyzed by flow cytometry. Mouse tissues were digested using collagenase. Flow cytometry was performed on a BD LSRFortessa. Endothelial cells were gated as CD45-CD31+PLVAP+. Data were analyzed using FCS Express software.

Primary antibodies used in flow cytometry included rat anti-mouse CD31 (clone 390, eBioscience), rat anti-mouse CD45 (clone 30-F11, BioLegend), and rat anti-mouse PV-1 (clone MECA-32, BioLegend). Mice were infected intranasally with influenza virus. Lung tissues were processed into single-cell suspensions. Flow cytometry was conducted using a BD LSR II. Endothelial cell subsets were identified based on CD31 and PV-1 expression.

PV-1, a marker of fenestrated endothelium, was stained using rat anti-mouse PV-1 (clone MECA-32) in flow cytometry experiments. Fenestrated endothelial cells were isolated from mouse kidneys. Cells were stained for CD31, VE-cadherin, and PV-1. Flow cytometry was performed on a FACSCalibur. Permeability assays were conducted in vivo using tracer dyes.

Caveolae, fenestrae and transendothelial channels retain PV1 on the surface of endothelial cells.

To verify the presence of PV1 on the surface of endothelial cells, cells were stained with a rat monoclonal antibody against PV1 (clone MECA-32; BD Pharmingen) and analyzed by flow cytometry. Endothelial cells were isolated from mouse lungs. Cells were stained with anti-CD31 and anti-PV1 antibodies. Flow cytometry was performed using a BD FACSAria. Surface expression was quantified relative to isotype controls.
