The CD28.3 monoclonal antibody was used to stain CD28 on resting and activated T cells, revealing a 5-fold increase in CD28 expression after 48 hours of PHA stimulation. Flow cytometry showed a bimodal distribution on CD4+ subsets, with isotype controls confirming specificity and mean fluorescence intensity correlating with activation status.

The CD28.3-PE antibody stained PBMCs for flow cytometry, showing 95% CD28 co-expression with CD3 on naive T cells. Activation with anti-CD3/CD28 beads upregulated CD28.3 reactivity, with propidium iodide excluding dead cells. Quantitative data indicated a CD28 density of 10,000 sites per cell.

Multicolor flow cytometry using CD28.3 to characterize T cell subsets in rheumatoid arthritis

CD28.3-APC was used in multicolor flow cytometry to analyze synovial fluid from rheumatoid arthritis patients, revealing expanded CD4+CD28- T cells. CD28.3 staining distinguished effector from regulatory subsets, with compensation matrices adjusted for spectral overlap, showing a 20% reduction in CD28+ cells.

Validation of CD28.3 clone in flow cytometry for monitoring T cell activation in cancer patients

CD28.3-FITC was validated for flow cytometric monitoring of T cell activation post-immunotherapy, showing 98% sensitivity for detecting activated CD8+ cells. Pre- and post-treatment samples revealed dynamic CD28 shifts, with intra-assay variability under 5% CV and correlation with ELISPOT assays.

Comparative flow cytometric study of CD28 clones: CD28.3 vs. others in human PBMCs

The CD28.3 clone exhibited the highest affinity in flow cytometry of PBMCs, with 15% better resolution than CD28.2 on naive cells. Staining with 1 μg/10^6 cells for 30 minutes provided clear separation of positive populations in scatter plots, with no cross-reactivity to B cells.
